细胞外基质糖基化对表皮干细胞生物学特性的影响

【摘要】: [目的]糖尿病是日常生活常见疾病之一,糖尿病下肢溃疡是其并发症之一,且经久难愈,因此糖尿病难愈创面成为人们研究的热点。表皮干细胞作为皮肤发生修复重建的关键源泉,糖尿病创面难愈是否与此有关呢?本实验通过探讨细胞基质糖基化对表皮干细胞黏附、增殖、凋亡等生物学行为的影响,进一步寻找糖尿病创面难愈机制。 [方法]利用Ⅳ胶原的快速黏附法分离培养表皮干细胞。然后利用免疫细胞化学和免疫荧光化学分别对角蛋白19和β1整合素进行鉴别。制作并鉴定糖基化Ⅳ胶原,将制备的表皮细胞悬液分别接种在包被有正常Ⅳ胶原和糖基化Ⅳ胶原(胶原糖基化7天,14天,21天)的6孔板上。15分钟后,每个组取3个孔,洗去未黏附细胞,分别计算其黏附率并进行比较;取第2代表皮干细胞,以5×10~3接种于预先包被有正常Ⅳ胶原和糖基化Ⅳ胶原的96孔板内,分别取培养3小时、2天、4天、6天、8天、10天、12天、14天细胞每天取5孔用MTT比色法测OD值,取其平均值并绘制生长曲线图;取第2代成对数生长期的糖基化组和正常胶原组细胞,按200/孔接种6孔板,2周之后测定克隆生成率;表皮干细胞按1×10~6/孔接种于包被有正常胶原和糖基化胶原6孔板上,培养6天和10天后流式细胞仪检测Annexin V Fitc比较两者细胞凋亡情况。 [结果]培养出来的表皮干细胞通过鉴定角蛋白19和β1整合素均有阳性表达;Ⅳ胶原糖基化7天,糖基化Ⅳ胶原的荧光值为1.864±0.126,而此时正常未进行糖基化的Ⅳ胶原荧光值为0.699±0.0831,糖基化14天,糖基化Ⅳ胶原的荧光值为3.051±0.112,而正常Ⅳ胶原荧光值为0.765±0.0927;糖基化21天,糖基化Ⅳ胶原的荧光值为4.416±0.1378,而正常Ⅳ胶原荧光值为0.8254±0.058;胶原在高糖下孵育7天时,糖基化组与正常胶原组黏附率没差异,P值>0.05;胶原在高糖下孵育14天时糖基化组比正常组黏附率低,P值<0.05,21天时糖基化组比正常组黏附率显著降低,P值<0.01;可见随着糖基化程度的加深,对表皮干细胞的黏附率逐渐降低;通过MTT绘制表皮干细胞在正常Ⅳ胶原和糖基化Ⅳ胶原上的生长曲线,可以看出在4天前细胞增殖缓慢,两者间没有差异,6天后开始出现差异,正常胶原组细胞增长速度要快于糖基化组;表皮干细胞在正常Ⅳ胶原和糖基化Ⅳ胶原上的克隆生成率比较:糖基化组为(38.5±1.9)%,正常胶原组为(32.7±1.5)%,糖基化组克隆生成率低于正常组胶原组。P<0.05;表皮干细胞在正常Ⅳ胶原和糖基化Ⅳ胶原上的凋亡比较:细胞孵育6天时,正常胶原组Annexin V阳性细胞数为(10.7±0.9438)%,而糖基化胶原组为(16.22±0.7912)%;细胞孵育第10天时正常胶原组Annexin V阳性细胞数为(4.66±0.7368)%,而糖基化胶原组为(8.266±0.8177)%。糖基化胶原组较正常胶原组Annexin V阳性细胞数明显增多。P<0.05 [结论]基质糖基化抑制了表皮干细胞的黏附和增殖,促进了表皮干细胞的凋亡,可能是糖尿病创面难愈机制之一。
