Fas系统在睾酮所诱导的小鼠骨髓巨噬细胞凋亡中的作用

细胞凋亡与组织器官的发育及分化、免疫细胞的发生及功能的维持、肿瘤的发生及转归等均有密切的关系,细胞以凋亡的方式主动“自杀”对维持组织的自身稳定具有积极意义。巨噬细胞作为一种重要的免疫效应细胞,其在机体免疫中的作用不可忽视,巨噬细胞的正常凋亡对于其正常行使功能有着重要的意义。影响巨噬细胞凋亡的因素有很多,其中性激素是不可忽视的一个因素。我们以前的研究表明,睾酮能够抑制感染了杜氏利什曼原虫的小鼠骨髓巨噬细胞的凋亡,那么,睾酮直接作用于小鼠骨髓巨噬细胞时,对其调亡的影响如何呢?此外,已知Fas及FasL是介导细胞凋亡的一对膜蛋白,当FasL与Fas结合时,可在数小时内诱发Fas表达细胞凋亡,且巨噬细胞上有Fas的表达;并已知NO是存在于体内有多种生物学活性的无机分子,它可以通过影响细胞物质代谢和能量代谢而造成细胞死亡,且有报道认为NO是诱导巨噬细胞凋亡的因子之一,故本研究的目的是观察在睾酮的直接作用下,小鼠骨髓巨噬细胞会否发生凋亡;如果发生凋亡,那么在这个过程中,Fas系统和NO是否参与了作用。 实验方法: 1.动物M乏一5 ter Work e57BI丫6j小鼠购自,},...
