转录抑制LPL表达的ZHX2蛋白功能域的鉴定

ZHX2(zinc fingers and homoboxes 2),即锌指结构与同源结构域蛋白2,是一个转录抑制因子,与ZHX1、ZHX3组成锌指结构及同源结构域家族蛋白。研究发现,ZHX2在肝癌、肺癌、霍奇金淋巴瘤等多种恶性肿瘤疾病中表达下调,因此ZHX2被鉴定为一个新的抑癌基因。此外,ZHX2尚可转录抑制肝癌细胞中重要的肿瘤标记物甲胎蛋白(AFP)、GPC3的表达,并通过调控细胞周期蛋白,抑制细胞的过度增殖,进一步说明ZHX2在抑制肿瘤发生和进程中起着重要作用,并为肝癌的诊断和预后监测提供了新的候选分子。与其他遗传背景相似的小鼠相比,Balb/cJ小鼠肝脏内的ZHX2表达水平显著降低,并可以避免高脂饮食饲喂诱导的高脂血症。通过数量性状位点图谱鉴定发现,脂质代谢相关基因LPL的表达水平与ZHX2表达水平呈现相反的表达趋势,这也许是因为饲喂高脂饮食后,Balb/cJ小鼠肝脏中高表达的LPL能摄取更多血浆甘油三酯,使血浆中甘油三酯的水平低于其他品系的小鼠。LPL全称脂蛋白脂肪酶,是机体分解甘油三酯的关键酶。LPL通过肝素结合位点以同源二聚体的形式锚定在血管内皮细胞表面,负责摄取载脂蛋白...
