幼年特发性关节炎患儿关节滑膜组织中趋化因子CXCL10及其受体CXCR3的表达及意义

【摘要】:目的检测趋化因子CXCL10及其受体CXCR3在幼年特发性关节炎(JIA)患儿关节滑膜组织中的表达,探讨其在JIA发病机制中的作用。方法通过免疫组织化学方法,检测12例JIA患儿和4例非JIA患儿关节滑膜组织中CXCL10和CXCR3的表达;通过半定量RT-PCR方法,检测CXCR3在JIA患儿和对照幼儿关节滑膜组织中mRNA水平的表达。结果免疫组织化学结果表明,CXCL10/CXCR3在JIA患儿及对照组阳性表达差异有显著性(P0.05);CXCR3在JIA患儿关节滑膜组织中mRNA表达水平(CXCR3∶GAPDH为2.26±1.55)显著高于对照组(0.66±0.44),两者差异有显著性(P0.05)。结论趋化因子CXCL10及其受体CXCR3在幼年特发性关节炎(JIA)的发病中可能起到了重要的作用。
[Abstract]:Objective to detect the expression of chemokine CXCL10 and its receptor CXCR3 in synovium of children with juvenile idiopathic arthritis (JIA) and to explore the role of them in the pathogenesis of JIA. The expression of CXCL10 and CXCR3 in synovial tissues of 12 children with JIA and 4 children with non JIA were detected, and the expression of CXCL10 and CXCR3 in synovial tissues of 12 children with JIA and 4 children with non JIA were detected. The expression of CXCR3 in synovial tissue of children with JIA and control was detected. Results the positive expression of CXCL10 / CXCR3 in children with JIA and control group was significantly higher than that in control group. There was a significant difference in mRNA expression in synovial tissue of children with JIA. The expression level of CXCR3: GAPDH was 2.26 卤1.55, which was significantly higher than that of control group (0.66 卤0.44). Conclusion the chemokine CXCL10 and its receptor CXCR3 may play an important role in the pathogenesis of juvenile idiopathic arthritis.
