血红素加氧酶-1在高氧肺损伤小鼠中的表达及作用

【摘要】:目的探讨血红素加氧酶-1(HO-1)在高氧造成的肺损伤转基因小鼠和正常小鼠中的肺部表达水平及其作用。方法把32只新生小鼠分成4组:野生型组(WT组),特异性转基因小鼠全身高水平表达HO-1组[HO-1-FL(H)组,细胞质]、特异性转基因小鼠全身低水平表达HO-1组[HO-1-FL(L)组,细胞质]和切去C末端HO-1(Nuc-HO-1-TR组,细胞核)。把4组小鼠暴露在高氧环境中3d,然后放到正常空气环境中,通过免疫组织化学、免疫荧光等实验技术来观察小鼠3d、7d和14d肺泡发育情况及HO-1在小鼠肺部表达情况。结果 HO-1在HO-1-FL(H)组小鼠肺部高表达,高氧暴露3d后4组小鼠肺部肺泡发育都受到了损害,在正常空气环境中7、14d时HO-1-FL(H)组小鼠的肺泡发育恢复得最好。结论小鼠肺部适度高水平的HO-1表达有助于高氧环境造成的小鼠肺损伤的恢复。
[Abstract]:Objective to investigate the lung expression of heme oxygenase-1 (HO-1) in hyperoxia-induced lung injury transgenic mice and normal mice. Methods 32 newborn mice were divided into 4 groups: wild type group, WT group, and specific transgenic group. High level expression of HO-1 group [HO-1-FLH] group, Specific transgenic mice expressed low level of HO-1 group [HO-1-FLL group, cytoplasm] and C terminal HO-1(Nuc-HO-1-TR group, nucleus. 4 groups of mice were exposed to hyperoxic environment for 3 days, then put into normal air environment, through immunohistochemistry, 4 groups of mice were exposed to hyperoxia for 3 days. Immunofluorescence technique was used to observe the alveolar development and the expression of HO-1 in the lungs of mice on day 7 and 14. Results the expression of HO-1 in the lungs of HO-1-FLH) group was high, and the alveolar development of the four groups were damaged after 3 days of hyperoxia exposure. The alveolar development of HO-1-FLH) group was the best in normal air environment for 14 days. Conclusion the moderate and high level of HO-1 expression in the lung of mice is helpful to the recovery of lung injury induced by hyperoxia.
