皮质酮对新生大鼠神经元迁移蛋白DCX、LIS1表达的影响

【摘要】:目的研究皮质酮(corticosterone)对新生大鼠大脑神经元迁移蛋白DCX(doublecortin、LIS1(microtubule binding lissencephaly1)表达的影响。 方法将48只生后(postnatal, PN)7d的Sprague-Dawley大鼠随机分成对照组和皮质酮干预组,皮质酮干预组于每日晨起(8时-9时)腹腔注射皮质酮(1.5mg/kg),每日一次,连续6天;对照组腹腔注射等量生理盐水。每组分别于PN14d、17d和21d选取8只断头取脑,用Western Blot技术及免疫组织化学染色观察大鼠大脑皮层及海马DCX、LIS1表达的变化。 结果PN14d、17d和21d时,大脑皮层及海马CA1-2、CA3-4、DG区皮质酮干预组DCX免疫组化累计光密度(AOD)均明显高于正常对照组(P㩳.05): Western Blot示皮质酮干预组海马区DCX蛋白表达明显高于正常对照组(P0.05)。与对照组比较,日龄14天大脑皮层及海马CA1-2、CA3-4、DG区皮质酮干预组LIS1免疫组化AOD均明显低于正常对照组(P0.05);日龄14天、21天皮层及日龄21天海马皮质酮干预组LIS1蛋白表达明显低于正常对照组(P0.05)。 结论1.LIS1、DCX在正常新生大鼠大脑中表达随时间进展表现出一定的规律性。2.外源性糖皮质激素影响神经移行相关蛋白DCX和LIS1表达,可能参与到GC对脑发育的影响中。
[Abstract]:Objective to study the effect of corticosterone on the expression of brain neuron migration protein DCX doublecortin LIS1 microtubule binding lissencephaly1 in neonatal rats.Methods 48 postnatal Sprague-Dawley rats of postnatal PN)7d were randomly divided into control group and corticosterone intervention group. In the corticosterone intervention group, 1.5 mg / kg corticosterone was injected intraperitoneally from 8 to 9 am daily for 6 days, while the control group was intraperitoneally injected with the same amount of normal saline.In each group, 8 decapitated rats were selected at 14 d and 21 d, respectively. The changes of the expression of DCXT LIS1 in rat cerebral cortex and hippocampus were observed by Western Blot technique and immunohistochemical staining.Compared with the control group,The immunohistochemical AOD of LIS1 in cortical cortex and hippocampal CA1-2CA3-4DG intervention group was significantly lower than that in normal control group (P 0.05), and the expression of LIS1 protein in cortex and hippocampal corticosterone intervention group was significantly lower than that in normal control group.Conclusion 1. The expression of LIS1 and DCX in the brain of normal neonatal rats showed a certain regularity over time. 2.Exogenous glucocorticoids affect the expression of DCX and LIS1, which may be involved in the effects of GC on brain development.
