金属蛋白酶7及金属蛋白酶19在胆道闭锁幼鼠动物模型肝脏组织中的表达

【摘要】:目的检测金属蛋白酶家族中金属蛋白酶7(MMP-7)和金属蛋白酶19(MMP-19)在幼鼠胆道闭锁动物模型中的表达情况,探讨其在胆道闭锁肝纤维化中的作用及机制。方法选取出生3 d的Wistar幼鼠132只作为实验组,采用肝外胆管结扎的方法制作胆道闭锁动物模型,62只幼鼠作为对照组,仅行开腹探查术。于术后3 d、7 d、14 d、21 d、28 d取肝脏组织,应用免疫组化、Western blotting及实时PCR等方法分别检测MMP-7及MMP-19在2组肝脏组织中的分布和表达情况。结果实验组幼鼠在术后24h即表现出梗阻性黄疸的临床症状;HE染色及Masson染色均发现肝脏组织纤维化逐渐加重;免疫组化、Western blotting、实时PCR等均发现实验组MMP-7的表达明显高于对照组,且逐渐升高;MMP-19在各时间段表达逐渐增强,但均低于对照组。结论 MMP-7和MMP-19在胆道闭锁肝纤维化中可能发挥重要作用。
[Abstract]:Objective to investigate the expression of metalloproteinase 7 (MMP-7) and metalloproteinase 19 (MMP-19) in the biliary atresia model of young rats and to explore the role and mechanism of metalloproteinase 7 (MMP-7) and metalloproteinase 19 (MMP-19) in hepatic fibrosis of biliary atresia. Methods 132 young Wistar rats were selected as experimental group, and 62 young rats were used as control group to make biliary atresia model by ligation of extrahepatic bile duct. Liver tissues were taken at 3 days, 7 days, 14 days, 21 days and 28 days after operation. The distribution and expression of MMP-7 and MMP-19 in the liver tissues of the two groups were detected by immunohistochemical, Western blotting and real-time PCR methods. Results the young rats in the experimental group showed the clinical symptoms of obstructive jaundice 24 hours after operation, and liver fibrosis was gradually aggravated by HE staining and Masson staining. The expression of MMP-7 in the experimental group was significantly higher than that in the control group, and the expression of MMP-19 was gradually increased, but lower than that in the control group. Conclusion MMP-7 and MMP-19 may play an important role in biliary atresia liver fibrosis.
