SARM抑制TRIF在RSV感染后气道炎症及气道高反应性作用机制研究

第一部分RSV感染后SARM及TRIF表达水平研究目的：RSV是引起5岁以下儿童下呼吸道感染最常见的病毒病原体。Toll样受体（TLR）信号通路与RSV致病机制密切相关。RSV感染后可通过激活TLR-TRIF依赖的信号通路引起气道炎症及气道高反应性。SARM为TRIF的负性调节因子，对RSV感染所致的气道炎症及气道高反应性（AHR）影响作用尚不清楚。本实验目的是研究RSV感染后SARM及TRIF的表达情况。方法：体外培养人类气道上皮细胞株9HTEo，RSV以MOI=10感染9HTEo细胞，分别在12h、24h、36h、48h和72h后收集细胞，提取全蛋白，Western blot方法检测感染后不同时间点SARM及TRIF的水平变化。RSV或紫外线灭活的RSV （UV RSV）感染6～8周龄雌性BALB/c小鼠感染，分别在感染的第3、5、7天处死，取小鼠肺组织，提取肺组织总蛋白，Western blot方法检测SARM及TRIF的变化情况。利用RES进行进一步干预，将6～8周龄雌性BALB/c小鼠分为对照组、RSV+PBS组、RSV+RES组，每组各5只，感染后第5天Western bl...
