SARM抑制TRIF在RSV感染后气道炎症及气道高反应性作用机制研究

第一部分RSV感染后SARM及TRIF表达水平研究目的:RSV是引起5岁以下儿童下呼吸道感染最常见的病毒病原体。Toll样受体(TLR)信号通路与RSV致病机制密切相关。RSV感染后可通过激活TLR-TRIF依赖的信号通路引起气道炎症及气道高反应性。SARM为TRIF的负性调节因子,对RSV感染所致的气道炎症及气道高反应性(AHR)影响作用尚不清楚。本实验目的是研究RSV感染后SARM及TRIF的表达情况。方法:体外培养人类气道上皮细胞株9HTEo,RSV以MOI=10感染9HTEo细胞,分别在12h、24h、36h、48h和72h后收集细胞,提取全蛋白,Western blot方法检测感染后不同时间点SARM及TRIF的水平变化。RSV或紫外线灭活的RSV (UV RSV)感染6~8周龄雌性BALB/c小鼠感染,分别在感染的第3、5、7天处死,取小鼠肺组织,提取肺组织总蛋白,Western blot方法检测SARM及TRIF的变化情况。利用RES进行进一步干预,将6~8周龄雌性BALB/c小鼠分为对照组、RSV+PBS组、RSV+RES组,每组各5只,感染后第5天Western bl...
