促红细胞生成素与黄芪对新生儿窒息后血清诱导人肾小管细胞TLR 4 /MyD88表达的影响

目的:研究促红细胞生成素与黄芪对新生儿窒息后血清诱导人肾小管细胞TLR4/MyD88 (Toll-like receptors 4/myeloid differentiation factor 88)表达的影响方法:(1)以HK-2细胞(Human renal proximal tubular cell,人近曲肾小管上皮细胞)作为研究对象。(2)以足月重度窒息(生后Apgar评分均小于4分)新生儿第一个24小时血清(20%体积分数)作为致病因素。(3)实验分四组,即对照组、窒息组、EPO(erythropoietin,促红细胞生成素)组、黄芪(Astragalus, AS)组。(4)检测指标:倒置相差显微镜下观察HK-2细胞形态变化;流式细胞仪(Flow Cytometry, FCM)检测各组细胞TLR4特异性荧光抗体阳性结合率表达情况;采用免疫组织化学法(SP法)检测各组细胞MyD88表达情况。(5)统计学分析:数据均用x±s表示,采用one-way ANOVA分析,组间比较用LSD法,运用SPSS14.0软件进行统计学处理,P<0.05为差异具有统计学意义。结果:(1)倒置相...
