【摘要】:目的:探讨fibulin糖蛋白家族中fibulin-3(EFEMP1)对卵巢癌细胞增殖、侵袭与转移的影响,为卵巢癌针对性治疗提供理论基础。方法:利用慢病毒转染RNA干扰的方式,转染具有高侵袭转移能力的S1克隆细胞株,抑制EFEMP1表达。Western blot、Real-time RT-PCR和免疫组化对转染效果进行验证。采用体外细胞功能分析技术检测干扰前后EFEMP1对细胞增殖、侵袭与转移能力的影响。建立裸鼠皮下移植瘤模型,体内观察EFEMP1对肿瘤生长的影响。结果:RNA干扰有效抑制EFEMP1表达,EFEMP1降表达显著抑制肿瘤细胞增殖、侵袭和转移能力,裸鼠体内成瘤率降低并且肿瘤体积显著下降,同时肿瘤组织Fibulin基因表达显著降低。结论:EFEMP1与卵巢癌发生发展及侵袭转移密切相关,为卵巢癌的治疗提供新的思路。
[Abstract]:Objective: to investigate the effect of fibulin-3 (EFEMP1) in fibulin glycoprotein family on the proliferation, invasion and metastasis of ovarian cancer cells. Methods: using lentivirus transfection of RNA interference to transfect S1 clone cell line with high invasion and metastasis ability. Inhibition of EFEMP1 expression. Western blot. Real-time RT-PCR and immunohistochemistry were used to verify the transfection effect. In vitro cell function analysis was used to detect the proliferation of EFEMP1 before and after interference. The effect of EFEMP1 on tumor growth was observed in vivo. Results EFEMP1 interference could effectively inhibit the expression of EFEMP1. The down-regulated expression of EFEMP1 significantly inhibited the proliferation, invasion and metastasis of tumor cells, and the tumorigenesis rate and tumor volume in nude mice decreased significantly. At the same time, the expression of Fibulin gene in tumor tissue was significantly decreased. Conclusion the expression of Fibulin gene is closely related to the occurrence, development, invasion and metastasis of ovarian cancer, which provides a new idea for the treatment of ovarian cancer.
