【摘要】:目的:寻找上皮性卵巢癌(epithelial ovarian cancer,EOC)癌相关成纤维细胞(cancer associated fibroblasts,CAFs)和正常成纤维细胞(normal fibroblasts,NFs)中差异表达的m RNA并进行分析,为进一步阐明肿瘤间质中癌相关成纤维细胞影响卵巢癌细胞恶性行为的机制提供依据。方法:分别提取3例上皮性卵巢癌的CAFs及3例正常卵巢组织的NFs,抽提总RNA,进行全基因表达谱分析。对差异基因进行基因本体论(Gene Ontology,GO)富集及信号通路联合分析,推定相关差异基因生物学功能,并对MDM2、NR3C1等6个上调的差异基因进行荧光定量PCR验证。结果:对芯片结果的分析显示差异基因共1 636个。q RTPCR对MDM2、NR3C1等6个差异基因的验证结果与芯片结果相符,生物信息学分析显示上述差异基因与癌相关通路、肌动蛋白细胞骨架重塑通路等有关,提示可能参与了对卵巢癌细胞恶性行为的促进作用。结论:EOC肿瘤间质中的成纤维细胞存在调控癌细胞的差异基因,分析这些差异基因为进一步研究卵巢癌成纤维细胞调控癌细胞生物学行为的分子机制提供了基本依据。
[Abstract]:Objective: to search for epithelial ovarian cancer (epithelial ovarian, cancer, EOC) cancer associated fibroblasts (cancer associated, fibroblasts, CAFs) and normal fibroblasts (normal, fibroblasts, NFs) in the differential expression of M RNA and analysis, provides the basis for further clarify the tumor stroma in cancer related to the malignant behavior of ovarian cancer cells the mechanism of the influence of fiber cells. Methods: CAFs were extracted from 3 cases of epithelial ovarian cancer and 3 cases of normal ovarian tissue NFs, extract the total RNA spectrum to analyze the gene expression of different genes. Gene Ontology (Gene Ontology, GO) and signaling pathway enrichment analysis, correlation between the putative gene biological functions and, MDM2, fluorescence quantitative PCR validation of differential gene NR3C1 6 up-regulated. Results: analysis of the microarray results showed differences in 1636 genes of.Q RTPCR MDM2, gene NR3C1 6 differences The results are consistent with the microarray results, bioinformatics analysis showed that the difference of gene and cancer related pathways, actin cytoskeleton remodeling pathway, suggesting that it may be involved in the malignant behavior of ovarian cancer cells. Conclusion: EOC tumor interstitial fibroblast gene is differentially regulated in cancer cells, analysis the basis for these differential genes for further research of the molecular mechanism of ovarian cancer cells into fibroblasts regulates the biological behavior of cancer.
