γ-神经突触核蛋白基因沉默对子宫内膜癌HEC-1A细胞周期的影响

【摘要】:目的观察γ-神经突触核蛋白(SNCG)基因稳定沉默后人子宫内膜癌HEC-1A细胞生长周期的变化。方法采用流式细胞术检测SNCG基因沉默后HEC-1A细胞周期变化;通过激光扫描共聚焦显微镜观察吖啶橙染色后G2/M期百分比变化。结果流式细胞术结果显示,SNCG基因沉默后HEC-1A细胞处于G_1和G_2/M期增多,处于S期减少,差异有统计学意义(P0.05);共聚焦显微镜观察结果显示,SNCG基因沉默后HEC-1A细胞处于G_2/M期百分比增高,差异有统计学意义(P0.05)。结论两个方法共同验证了慢病毒质粒载体shRNA干扰后SNCG基因稳定沉默的HEC-1A细胞周期阻滞于G_2/M期,提示SNCG基因与子宫内膜癌细胞生长周期密切相关。
[Abstract]:Objective to observe the expression of 纬 -neurosynaptophysin (SNCGG). Methods the changes of HEC-1A cell cycle after SNCG gene silencing were detected by flow cytometry. The percentage change of G _ 2 / M phase after acridine orange staining was observed by laser scanning confocal microscope. After SNCG gene silencing, the number of HEC-1A cells increased in Gsta1 and G2 / M phase, and decreased in S phase. The difference was statistically significant (P 0.05). The results of confocal microscopy showed that the percentage of HEC-1A cells in 2 / M phase was increased after the silencing of SNCG gene. The difference was statistically significant (P0.05). Conclusion the two methods combined to verify that the stable silencing HEC-1A cell cycle of SNCG gene after the interference of lentivirus plasmid shRNA was blocked in G _ S _ 2 / M phase. The results suggest that SNCG gene is closely related to the growth cycle of endometrial cancer cells.
