卵泡刺激素受体启动子-29位点基因突变与卵巢反应性的关系

【摘要】:目的通过检测接受体外受精-胚胎移植(In vitro fertilization and embryo transfer, IVF-ET)治疗的患者外周血白细胞上卵泡刺激素受体(Follicle-stimulating Hormone Receptor, FSHR)启子-29位点的基因型,探讨该位点基因突变后表达改变与IVF-ET患者卵巢反应性的关系。 方法本研究根据在IVF-ET治疗周期中,患者在接受控制性超促排卵(Controlled ovarian hyperstimulation, COH)时卵巢对药物的反应性,将85例患者分为三组,其中卵巢高反应组20例,卵巢反应正常组46例,卵巢低反应组19例。采用聚合酶链式反应-限制性长度多态性技术(Polymerase chain reaction-restriction fragment length polymorphism, PCR-RFLP)来测定患者白细胞上FSHR启动子-29位点的基因型,并分析其与患者卵巢反应性的关系。 结果①卵巢低反应组FSHR启动子29位点G→A突变率为,42.10%,卵巢反应正常组的基因突变率为10.87%,卵巢高反应组的基因突变率为10%。 ②卵巢低反应组的基因突变率均高于卵巢反应正常组及高反应组(P0.05);卵巢反应正常组和高反应组的基因突变率比较无显著性差异(P0.05)。 ③卵巢低反应组HCG日雌二醇(estradiol, E,)值及荻卵数均低于其他两组(P0.05);卵巢反应正常组HCG (?) E值及获卵数均低于卵巢高反应组(P0.05)。 结论外周血白细胞FSHR启动子-29位点G→Λ基因突变可能与卵巢反应性不良有关,测定该位点的基因型对预测卵巢的反应性有一定的意义。
[Abstract]:Objective to detect in vitro fertilization and embryo transfer. Follicle-stimulating Hormone Receptor was obtained from peripheral white blood cells treated with IVF-ETS. To investigate the relationship between the gene mutation and ovarian reactivity in patients with IVF-ET. Methods according to the treatment cycle of IVF-ET, patients received controlled ovarian hyperstimulation with controlled hyperstimulation of ovulation. 85 patients were divided into three groups: ovarian hyperresponsiveness group (n = 20) and normal ovarian response group (n = 46). Methods Polymerase chain reaction (PCR) and restriction length Polymorphism (RFLP) technique were used in 19 cases of ovarian hyporesponse group (. Polymerase chain reaction-restriction fragment length polymorphism. PCR-RFLP was used to determine the genotype of FSHR promoter-29 on leukocytes and to analyze its relationship with ovarian reactivity. Results 1 FSHR promoter 29 locus G in ovarian hyporesponse group 鈫扵he mutation rate of A was 42.10, the mutation rate of normal ovary was 10.87, and the mutation rate of ovarian high response was 10. 2the gene mutation rate of ovarian low response group was higher than that of normal ovarian response group and high response group (P 0.05). There was no significant difference in gene mutation rate between normal ovarian response group and high response group (P 0.05). 3 the value of estradiol (E) and the number of eggs in the HCG group were lower than those in the other two groups (P 0.05), and the number of estradiol (E) in the normal ovarian response group was lower than that in the other two groups (P 0.05). The value of E and the number of oocytes obtained were lower than that of the ovarian hyperreactive group (P0.05A). Conclusion the FSHR promoter -29 in peripheral blood leukocytes is G 鈫扵he mutation of A gene may be related to ovarian reactivity, and the determination of the genotype of this locus may be useful in predicting ovarian reactivity.
