血浆miR-20a和let-7a在卵巢癌中的表达及其临床意义

【摘要】:目的检测miR-20a和let-7a在卵巢癌患者血浆中的表达,探讨其临床意义。方法选取卵巢癌患者50例为实验组,另选择同期与其年龄性别相匹配的健康体检者20例为健康对照组,采用实时定量荧光定量酶链反应(qRT-PCR)检测血浆miR-20a和let-7a表达水平,同时采用电化学发光法测定其血浆中糖类抗原125(CA125)的含量并进行统计分析。结果卵巢癌患者血浆中miR-20a和let-7a的相对表达量分别为(4.02±3.77)、(0.42±0.34),卵巢癌患者血浆中miR-20a和CA125的表达均高于对照组(P0.01),let-7a表达低于对照组(P0.01)。临床病理分期Ⅰ~Ⅱ期和Ⅲ~Ⅳ期卵巢癌患者血浆let-7a相对表达量的中位数分别为0.39、0.21,差异有统计学意义(P0.01),而血浆miR-20a的表达与临床病理分期无关(P0.05),miR-20a和let-7a的相对表达量与年龄、淋巴结转移和组织学分类均无关(P0.05)。miR-20a、let-7a和CA125的ROC曲线下面积为0.898(95%CI:0.802~0.957)、0.876(95%CI:0.775~0.942)、0.819(95%CI:0.708~0.900)。结论卵巢癌患者血浆中miR-20a和let-7a均表达异常,提示能够成为辅助诊断卵巢癌的标志物。
[Abstract]:Objective to detect the expression of miR-20a and let-7a in plasma of patients with ovarian cancer and to explore its clinical significance. Methods 50 patients with ovarian cancer were selected as experimental group and 20 healthy persons matched with their age and sex as healthy control group. The expression of miR-20a and let-7a in plasma was detected by real-time quantitative fluorescence quantitative polymerase chain reaction (qRT-PCR). The relative expression of miR-20a and let-7a in plasma of patients with ovarian cancer was 4.02 卤3.77 卤0.42 卤0.34, respectively. The expression of miR-20a and CA125 in plasma of patients with ovarian cancer was both determined by electrochemiluminescence.
