APLP2在子宫内膜癌中的表达及其对预后的预测价值

【摘要】:目的探讨子宫内膜癌中淀粉样肽前体样蛋白2(APLP2)的表达,分析其与各临床参数的相关性。方法通过数据挖掘,分析335例肿瘤基因图谱(TCGA)数据库中的子宫内膜癌RNA-SEQ数据并结合患者的完整生存期信息,采用Kaplan-Meier法分析APLP2表达与子宫内膜癌患者生存之间的关系。采用real-time PCR技术检测正常子宫内膜和子宫内膜癌各30例冰冻组织中APLP2 mRNA的表达情况。应用免疫组织化学法检测88例子宫内膜癌组织和32例正常子宫内膜组织中APLP2蛋白的表达。结果 Kaplan-Meier生存曲线分析表明,APLP2低表达的子宫内膜癌患者整体生存率明显降低(HR=2.87,P=0.002)。实时荧光定量PCR检测结果提示,APLP2 mRNA在子宫内膜癌组织中表达明显低于正常内膜组织(P=0.031)。免疫组织化学结果显示:APLP2蛋白在子宫内膜癌组织中的阳性表达率明显低于正常子宫内膜组织(P=0.025),其表达水平与雌激素受体表达水平相关(P=0.010)。结论 APLP2在子宫内膜癌中起到抑癌基因的功能。APLP2基因可能通过雌激素受体途径起作用。APLP2可能成为子宫内膜癌预后预测及治疗的靶点。
[Abstract]:Objective to investigate the expression of amyloid peptide precursor protein 2 (APLP2) in endometrial carcinoma and analyze its correlation with clinical parameters. To analyze the RNA-SEQ data of endometrial carcinoma in TCGA database of 335 cases of tumor, and combine the complete survival information of patients. The relationship between the expression of APLP2 and the survival of endometrial carcinoma was analyzed by Kaplan-Meier. The expression of APLP2 mRNA was detected by real-time PCR technique in 30 frozen tissues of normal endometrium and 30 cases of endometrial carcinoma. Immunohistochemistry was used to detect the expression of APLP2 mRNA in 30 cases of normal endometrium and 30 cases of endometrial carcinoma. The expression of APLP2 protein was detected in 88 cases of endometrial carcinoma and 32 cases of normal endometrial tissue. Results Kaplan-Meier survival curve analysis showed that the overall survival rate of endometrial carcinoma patients with low expression of APLP2 was significantly lower than that of normal endometrial tissues. Real-time fluorescence quantitative analysis showed that the overall survival rate of patients with endometrial carcinoma with low expression of APLP2 was significantly lower than that of patients with endometrial carcinoma. The expression of APLP2 mRNA in endometrial carcinoma tissues was significantly lower than that in normal endometrial tissues (P < 0.031). Immunohistochemical results showed that the positive expression rate of WAPLP2 protein in endometrial carcinoma was significantly lower than that in normal endometrial tissues. Conclusion APLP2 may play a role as a tumor suppressor gene in endometrial carcinoma. APLP2 gene may act through estrogen receptor pathway. APLP2 may be a prognostic preconditioning of endometrial carcinoma. The target of detection and treatment.
