妊娠早期稽留流产患者绒毛组织中HIF-1α、BNIP3、LC3的表达

【摘要】：目的:检测妊娠早期稽留流产患者绒毛组织中HIF-1α、BNIP3、LC3的表达,探讨滋养层细胞自噬异常与稽留流产的关系。方法:收集正常早孕妇女及稽留流产妇女绒毛组织各30例,采用实时荧光定量PCR法检测HIF-1α、BNIP3 mRNA的表达;采用免疫组化SP法及蛋白印迹法检测HIF-1α、BNIP3及LC3蛋白的表达。结果:两组绒毛组织中均可见HIF-1α、BNIP3及LC3蛋白的表达,HIF-1α主要定位于滋养细胞胞核及胞质,BNIP3及LC3主要定位于滋养层细胞胞质中。与正常早孕组相比,稽留流产组HIF-1α与BNIP3 mRNA和蛋白表达水平以及LC3Ⅱ/Ⅰ均明显下降(P0.05)。结论:绒毛组织滋养层细胞中HIF-1α/BNIP3信号通路调节的低氧诱导自噬水平下降可能是稽留流产发生的重要机制。
[Abstract]:Objective: to detect the expression of HIF-1 伪 BNIP3LC3 in the chorionic villi of patients with missed abortion in early pregnancy, and to explore the relationship between abnormal autophagy of trophoblastic cells and missed abortion. Methods: 30 cases of normal early pregnant women and 30 cases of women with missed abortion were collected. The expression of HIF-1 伪 -BNIP3 mRNA was detected by real-time fluorescence quantitative PCR.
