输卵管:间充质干细胞的新来源

【摘要】:目的:探讨临床上废弃的输卵管组织中是否存在间充质干细胞(MSC)。方法:收取2013年6月至9月在北京海军总医院手术治疗的6例因子宫颈癌、子宫肌瘤、子宫内膜癌及卵巢癌等疾病行输卵管切除术的输卵管组织,体外细胞培养、流式细胞学及体外诱导分化鉴定此类细胞为输卵管MSC。结果:体外分离、培养及扩增的输卵管细胞形态呈长梭型,细胞排列均一并呈漩涡状生长。流式细胞术检测结果显示,≥95%的MSC表达MSC相关表面标记物CD105、CD73和CD90,CD13、CD29、CD44、CD166、HLA-ABC均阳性表达,CD14、CD19、CD34、CD45、HLA-DR均阴性表达。体外诱导分化显示,输卵管干细胞具有成骨、成脂肪及成软骨分化的能力;RT-PCR结果显示,输卵管干细胞经诱导分化后,成脂相关基因AP2,成骨相关基因Osteopontin及成软骨相关基因Sox-9和COMP表达均阳性。结论:临床废弃的输卵管组织中存在MSC,提示临床中废弃的输卵管组织是MSC的新来源。
[Abstract]:Objective: to investigate the existence of mesenchymal stem cells (MSCs) in the discarded oviduct tissue. Methods: from June 2013 to September, 6 cases of cervical cancer and uterine leiomyoma were treated in Beijing Naval General Hospital. The oviduct tissue of endometrial carcinoma and ovarian cancer underwent salpingotomy in vitro cell culture flow cytology and differentiation in vitro were used to identify such cells as fallopian tube MSC.Results: in vitro isolation. The cultured and expanded tubal cells showed long fusiform, uniform arrangement and whirlpool growth. The results of flow cytometry showed that, The expression of MSC related surface markers CD105, CD73 and CD90, CD13, CD29, CD44, CD46, CD166T, and HLA-DR were negative in MSC 鈮,
