大鼠双侧卵巢切除术后不同时期血清骨生化标记物与骨小梁三维结构变化之间的研究

【摘要】:目的观察双侧卵巢切除(OVX) SD大鼠不同时间点血清骨转换标记物、骨小梁三维结构动态变化。方法将32只6月龄雌性SD大鼠随机分为假手术(Sham)组和双侧卵巢切除术(OVX)组,在术后选择4个时间点(2、4、8、12周)随机选取样本,每个组别各时间点予以处死4只动物,抽取血清测量雌二醇(E2)、骨碱性磷酸酶(BALP)、抗酒石酸酸性磷酸酶-5b (TRACP-5b)浓度,侵入性手术取胫骨并用显微CT (Micro-CT)和显微镜HE染色方法观察胫骨骨小梁微结构变化。结果术后2周,OVX组E2较Sham组开始下降,在第4周到12周时有明显差异(P0.05),而Sham组E2水平随时间变化不大;OVX组血清BALP在术后2周到达峰值后呈现出下降趋势,在第12周时下降到术前的水平;而OVX组的血清TRACP-5B水平在术后迅速升高到第4周达峰值后出现下降趋势,但是在术后12周时让然大于Sham组及术前水平;用Micro-CT分析胫骨部位干骺端的相对骨体积(BV/TV)、骨小梁数目(Tb. N)、骨小梁分离度(Tb.Sp)、Conn. D、骨密度(BMD)在2周时与Sham组无明显差异,第4周开始出现明显差异,12周差异更加明显,骨小梁厚度(Tb. Th)始终无明显差异,即骨小梁BV/TV、Tb. N、Conn. D、BMD下降,Tb.Sp升高;Micro-CT重建显示术后12周OVX组骨小梁较Sham组稀疏,缺失严重;HE染色显示术后12周OVX组大鼠胫骨干骺端骨骺板下骨小梁变细且排列稀疏。结论OVX术骨质疏松模型建立过程中大鼠成骨及破骨细胞活跃度先增高后降低,破骨细胞相对而言上升周期更长且下降后能维持在更高水平,比成骨细胞更活跃,骨吸收大于骨生成导致骨质疏松,组织学检测结果表明骨小梁结构不成熟。
[Abstract]:Objective to observe the dynamic changes of serum osteoconversion markers and bone trabecular structure at different time points in bilateral ovariectomized OVX SD rats. Methods 32 6-month-old female SD rats were randomly divided into sham-operated group and bilateral ovariectomized group (OVX) group. Samples were randomly selected at 4 time points after operation. Four animals were killed at each time point in each group. Serum levels of estradiol E2P, bone alkaline phosphatase (BALPN), tartrate-resistant acid phosphatase (-5b) and TRACP-5b were measured. The microstructures of bone trabeculae were observed by microCT Micro-CTand HE staining in invasive operation. Results E2 in OVX group was lower than that in Sham group 2 weeks after operation. At the 4th week to 12th week, there was a significant difference (P 0.05), while the level of E 2 in Sham group showed a decreasing trend after reaching the peak value 2 weeks after operation, and decreased to the preoperative level at the 12th week. However, the level of serum TRACP-5B in OVX group increased rapidly to the peak at the 4th week after operation, and then decreased, but at 12 weeks after operation, it was higher than that in Sham group and preoperative level. Micro-CT was used to analyze the relative bone volume (BV / TVV), the number of trabeculae and bone trabeculae in the metaphyseal region of the tibia (BV / TVV), and the trabeculae separation degree Tb.Spn.Conn.D, bone density (BMD) were not significantly different from those in the Sham group at 2 weeks, but there were significant differences at the beginning of the 4th week and at the 12th week, the difference was even more obvious. There was no significant difference in bone trabecular thickness and Tb. Th. the trabecular bone trabeculae (BV / TVV Tb.NbConn.DUBMD) decreased and Tb.Sp was increased. Micro-CT reconstruction showed that trabecular bone in OVX group was sparse than that in Sham group 12 weeks after operation. The results of HE staining showed that the trabeculae of the tibial metaphyseal epiphyseal plate of the OVX group became thin and sparse 12 weeks after operation. Conclusion the osteogenesis and osteoclast activity of the osteoblasts and osteoclasts increased first and then decreased during the establishment of the osteoporosis model of OVX. Osteoclasts had a longer ascending cycle and decreased osteoclasts, which were more active than osteoblasts, and bone resorption was larger than osteogenesis, resulting in osteoporosis. Histological examination showed that the trabecular structure of bone was immature.
