【摘要】:目的本研究意在探讨SIX1蛋白在人宫颈癌组织中的表达情况及与宫颈癌生物学特性的关系。 方法所选实验标本例数共204例,组织芯片包括107例宫颈鳞癌,16例宫颈腺癌,另有32例正常宫颈、49例宫颈原位癌。应用免疫荧光染色法观察SIX1基因在HELA细胞系中的细胞定位情况,采用免疫组化SP法检测SIX1蛋白在各种宫颈组织中的表达情况,并分析SIX1蛋白的表达与宫颈癌临床病理学特征的关联。 结果1:免疫荧光染色发现:SIX1蛋白在宫颈癌HeLa细胞系中主要定位于细胞浆和核仁。 2:从正常宫颈上皮到CIN原位癌再到浸润癌,SIX1蛋白阳性表达量有显著升高趋势。与正常宫颈上皮组织相比,宫颈癌组织中SIX1蛋白阳性率及阳性强度明显升高,差异有统计学意义(P0.001)。 3:宫颈癌中SIX1蛋白的表达与肿瘤大小、淋巴转移、血液转移有关,有统计学差异(P0.05或P0.01),而与年龄、肿瘤分期以及肿瘤分化程度无关(P0.05)。 结论1:SIXl蛋白的过表达,可能参与了宫颈癌的发生发展过程。2.SIX1蛋白的表达与宫颈癌的生物学特性--侵袭和转移性行为有关。在临床工作中为宫颈癌的侵袭与转移提供了可能的分子标志物。
[Abstract]:Objective to investigate the expression of SIX1 protein in human cervical carcinoma and its relationship with the biological characteristics of cervical cancer.Methods A total of 204 samples were selected, including 107 cases of squamous cell carcinoma of the cervix and 16 cases of adenocarcinoma of the cervix, and there were 32 cases of normal cervical carcinoma in situ.The localization of SIX1 gene in HELA cell line was observed by immunofluorescence staining, and the expression of SIX1 protein in various cervical tissues was detected by immunohistochemical SP method.The relationship between the expression of SIX1 protein and clinicopathological features of cervical cancer was analyzed.Results 1. It was found by immunofluorescence staining that the protein was mainly located in cytoplasm and nucleoli in cervical cancer HeLa cell line.2: the positive expression of SIX 1 protein increased significantly from normal cervical epithelium to CIN carcinoma in situ and then to invasive carcinoma.Compared with normal cervical epithelium, the positive rate and intensity of SIX1 protein in cervical carcinoma tissue were significantly higher than those in normal cervical epithelium tissue (P 0.001).3. The expression of SIX1 protein in cervical cancer was related to tumor size, lymphatic metastasis and blood metastasis, there was significant difference (P0.05 or P0.01), but it was not related to age, tumor stage and tumor differentiation.Conclusion the overexpression of 1:SIXl protein may be involved in the genesis and development of cervical cancer. 2. The expression of SIX1 protein may be related to the biological characteristics of cervical cancer, invasion and metastasis.It provides a possible molecular marker for invasion and metastasis of cervical cancer.
