叶酸对宫颈癌SiHa及HeLa细胞自噬蛋白p62影响的初步研究

【摘要】:目的:探讨叶酸对宫颈癌Si Ha细胞及He La细胞自噬相关蛋白p62表达的影响。方法:在正常叶酸水平(正常对照组,1.0μg/ml)和低剂量叶酸水平(低剂量叶酸组,0.1μg/ml)两种条件下分别干预宫颈癌Si Ha细胞及He La细胞,并于24小时、48小时、72小时分别收取细胞,采用实时荧光定量PCR实验及免疫印迹实验分别检测自噬相关蛋白p62 m RNA水平及蛋白水平表达量的变化。结果:1.与正常对照组相比,低剂量叶酸组培养的宫颈癌Si Ha细胞,在蛋白水平其48小时、72小时的p62的表达量降低(P0.05),在m RNA水平仅72小时的p62表达量降低(P0.05)。2.与正常对照组相比,低剂量叶酸组培养的宫颈癌He La细胞,无论在蛋白水平还是m RNA水平,仅48小时的p62表达量降低(P0.05)。结论:1.低剂量叶酸使宫颈癌Si Ha细胞自噬相关蛋白p62的表达下降,提示低剂量叶酸可诱导宫颈癌细胞自噬的发生。2.低剂量叶酸使宫颈癌He La细胞自噬相关蛋白p62的表达下降,同样提示低剂量叶酸可诱导宫颈癌细胞自噬的发生。
[Abstract]:Aim: to investigate the effect of folic acid on the expression of autophagy associated protein p62 in Cervical carcinoma SIHA cells and He-La cells.Methods: normal folic acid (normal control group, 1.0 渭 g / ml) and low-dose folic acid (low dose folic acid group, 0.1 渭 g / ml) were used to treat cervical cancer cell line SIHA and He-La cells, respectively. Cells were collected at 24 hours, 48 hours and 72 hours, respectively.The level and expression of autophagy associated protein p62 m RNA were detected by real-time fluorescence quantitative PCR assay and Western blotting assay respectively.The result is 1: 1.Compared with the normal control group, the expression of p62 in the low dose folic acid group was decreased at the protein level at 48 hours and 72 hours, and the expression of p62 at m RNA level at 72 hours was decreased.Compared with the normal control group, the expression of p62 in He-La cells cultured with low dose folic acid at the protein level and m RNA level decreased only 48 hours.Conclusion 1.Low dose folic acid decreased the expression of autophagy associated protein p62 in Cervical carcinoma SIHA cells, suggesting that low dose folic acid could induce the occurrence of autophagy in cervical cancer cells.Low dose folic acid decreased the expression of autophagy associated protein p62 in helium La cells, suggesting that low dose folic acid could induce autophagy in cervical cancer cells.
