母鼠注射海洛因对仔鼠大脑额叶结构及相关蛋白表达的影响

【摘要】:研究母鼠注射海洛因对仔鼠大脑额叶结构与相关蛋白表达的影响,选20只母鼠随机分为空白对照组和海洛因1,2,3组,从受孕第10d开始,各孕鼠分别腹腔连续注射生理盐水和2.0,4.0,6.0g·L-1的海洛因溶液,直至孕鼠分娩后15d,于仔鼠出生5d,10d和15d分别称量检测仔鼠大脑的重量,用光学显微镜观察其大脑额叶的显微结构变化,用免疫组化法检测额叶皮层NGF和Caspase-3蛋白的表达.结果表明,各海洛因组仔鼠大脑重量均显著或极显著低于空白对照组(P0.05或P0.01);海洛因组仔鼠额叶皮层组织结构有不同程度的发育缺陷,外颗粒层、内颗粒层、锥体细胞层和多形细胞层的细胞均出现空泡化和细胞凋亡现象,额叶皮层NGF表达增强,Caspase-3蛋白的表达极显著增强(P0.01).表明母鼠注射海洛因影响仔鼠大脑额叶组织的正常发育.
[Abstract]:To study the effect of heroin injection on the structure of frontal lobe and the expression of related proteins in the brain of young rats, 20 female rats were randomly divided into blank control group and heroin 1 + 2 + 3 group, starting from the 10th day of conception.Each pregnant rat was injected intraperitoneally with normal saline and 2.0 ~ 4.0 ~ 6.0 g 路L ~ (-1) heroin solution continuously until 15 days after delivery. The weight of the brain was measured on the 5th and 15th day after birth, and the microstructure of the frontal lobe of the newborn rat was observed by optical microscope.The expression of NGF and Caspase-3 in frontal cortex was detected by immunohistochemistry.The results showed that the brain weight of each heroin group was significantly lower than that of the blank control group (P0.05 or P0.01), and the structure of the frontal cortex of the heroin group had different degrees of developmental defects, including the outer granular layer and the inner granular layer.Vacuolation and apoptosis appeared in pyramidal cell layer and polymorphic cell layer, and the expression of Caspase-3 protein in frontal cortex increased significantly (P 0.01).The results showed that the injection of heroin affected the normal development of frontal lobe tissue.
