ΔNp63α基因过表达对人宫颈癌细胞SiHa基因表达谱的影响

【摘要】:目的分析ΔNp63α基因过表达时人宫颈癌细胞SiHa基因表达谱的变化,筛选受ΔNp63α调控的潜在靶基因。方法利用慢病毒转染技术构建稳定过表达ΔNp63α的转染组细胞株SiHa-ΔNp63α和对照组细胞株SiHa-NC。应用基因芯片技术研究两组细胞株基因表达谱变化,并进行生物学信息分析和实时荧光定量PCR验证。结果基因芯片数据共筛选出差异倍数1.5倍以上的基因共1 405个,其中843个基因在SiHa-ΔNp63α细胞中表达上调,562个表达下调。通过生物信息学分析显示这些基因主要参与细胞生长与周期、信号转导、细胞通讯、细胞黏附、细胞转移和侵袭等功能,涉及的信号通路包括抗原加工提呈、细胞因子与受体相互作用、细胞黏附分子、补体系统等。结论研究ΔNp63α调控的潜在靶基因或信号通路,对探索宫颈癌发生发展的机制有重要意义。
[Abstract]:Objective to analyze the changes of SiHa gene expression profile in human cervical cancer cells during overexpression of 螖 Np63 伪 gene, and to screen potential target genes regulated by 螖 Np63 伪.Methods the cell line SiHa- 螖 Np63 伪 of stable overexpression of 螖 Np63 伪 and the control cell line SiHa-NCwere constructed by lentivirus transfection technique.The gene expression profiles of two groups of cell lines were studied by gene chip technique, and the biological information analysis and real-time fluorescence quantitative PCR verification were performed.Results A total of 1 405 genes were screened by microarray data, in which 843 genes were up-regulated and down-regulated in SiHa- 螖 Np63 伪 cells.Bioinformatics analysis showed that these genes were mainly involved in cell growth and cell cycle, signal transduction, cell communication, cell adhesion, cell metastasis and invasion.Cytokines interact with receptors, cell adhesion molecules, complement systems, and so on.Conclusion it is important to study the potential target gene or signal pathway regulated by 螖 Np63 伪 in the pathogenesis of cervical cancer.
