褪黑素对SD大鼠雌性子代生长及性发育的影响

【摘要】:【目的】探讨孕期使用外源性褪黑素对子代生长发育的影响。【方法】SD孕鼠共30只,随机分成3组,每组10只。褪黑素低剂量组MT-LD组:给予褪黑素10 mg·kg-1·d-1,褪黑素高剂量组MT-HD组:40 mg·kg-1·d-1,对照组:生理盐水10 mg·kg-1·d-1处理。待分娩后选择每窝雌性子代每周测定其体质量平均增长情况。断奶后每日测定阴道开口及动情周期变化。于性周期稳定后测量每窝雌性子鼠股骨、胫骨的平均长度;ELISA方法检测血清褪黑素、生长激素(GH)、卵泡刺激素(FSH)、黄体生成素(LH)的平均水平,比较不同处理因素下每窝雌性子代体质量、长骨生长及性发育的差异。【结果】共获得30窝子鼠,分别为MT-LD 92只,MT-HD 84只,对照组97只。MT-HD组质量在各时间点均高于其余各组,MT-LD组平均体质量高于对照组(P0.01)。MT-HD组阴道开口时间及出现第一个动情间期时间较其余两组显著提前(P0.01),而MTLD与对照组间无明显差异。MT-HD组血清中FSH、LH、褪黑素、生长激素平均水平分别较其余两组高(P0.05),而上述各指标在MT-LD组及对照组见无统计学差异。【结论】外源性褪黑素作用于孕晚期母鼠,影响子代体质量、长骨生长及性发育,并存在剂量依赖关系,临床上在孕期使用褪黑素时应重视其对子代体质量、性发育及激素等的影响。
[Abstract]:[objective] to investigate the effects of exogenous melatonin on the growth and development of offspring during pregnancy. [methods] Thirty SD pregnant rats were randomly divided into 3 groups with 10 rats in each group.MT-LD group was treated with 10 mg kg-1 d -1 of melatonin, 10 mg kg-1 d -1 of MT-HD group with high dose of melatonin, and 10 mg kg-1 d -1 of normal saline in control group.After delivery, the average body mass of each litter of female offspring was measured weekly.The changes of vaginal opening and estrus cycle were measured daily after weaning.The mean length of femur and tibia were measured by Elisa to detect the average levels of serum melatonin (melatonin), growth hormone (GHH), follicle stimulating hormone (FSH) and luteinizing hormone (LH) in femur and tibia after sexual cycle stabilization.The weight, long bone growth and sexual development of female progenies in each litter were compared under different treatments. [results] A total of 30 litters of MT-LD 92 MT-HD 84 mice were obtained.The mean body mass of 97 MT-HD patients in the control group was higher than that in the other MT-LD group at all time points, and the vaginal opening time and the first estrous interval time in the MT-HD group were significantly earlier than those in the other two groups, while the average body mass in the MT-LD group was significantly higher than that in the control group, and between MTLD and the control group, the vaginal opening time and the first estrous interval time were significantly earlier.There was no significant difference in serum FSHLH and melatonin in MT-HD group.The average level of growth hormone was higher than that of the other two groups (P 0.05), but there was no statistical difference between the two groups. [conclusion] exogenous melatonin can affect the weight of offspring, the growth of long bone and the development of sex.The effect of melatonin on the weight, sex development and hormones of the offspring should be paid more attention to in clinical use of melatonin during pregnancy in a dose-dependent manner.
