自噬相关基因OPTN、P62与子宫内膜异位症的相关性研究

【摘要】:[目的]检测子宫内膜异位症(EMs)患者血清及异位子宫内膜、在位子宫内膜组织(卵巢子宫内膜异位囊肿)中自噬相关基因视神经病变诱导基因(Optineurin OPTN)、P62蛋白的表达,比较OPTN、P62在子宫内膜异位症患者与正常对照组中的不同表达,以了解OPTN、P62与子宫内膜异位症的关系,探讨自噬相关基因在子宫内膜异位症发病机制中的作用,为病因学寻找依据,为治疗寻找新的思路。[方法]1.运用酶联免疫法(Elisa)测定被研究者血清中OPTN、P62蛋白的表达。2.运用实时荧光定量PCR法(RT-PCR)测定EMs患者异位子宫内膜、在位子宫内膜组织及正常子宫内膜组织中OPTN、P62 mRNA的表达。3.运用Westren-Blot法测定EMs患者异位子宫内膜、在位子宫内膜组织及正常子宫内膜组织中OPTN、P62蛋白的表达。[结果]1.OPTN蛋白在EMs患者血清中的表达高于正常对照组血清中的表达水平(P0. 05) , P62蛋白在EMs患者血清中的表示低于正常对照组血清中的表达水平(P0. 05) 。2. OPTN mRNA在实验组的异位子宫内膜组织、在位子宫内膜组织中的表达均低于对照组子宫内膜(P0. 05) ; P62 mRNA在实验组的异位子宫内膜组织、在位子宫内膜组织中的表达高于对照组子宫内膜(P0. 05);但实验组的异位子宫内膜组织和在位子宫内膜组织中OPTN mRNA及P62 mRNA的表达为异位组低于在位组,差异无统计学意义(P0.05)。3.OPTN蛋白在实验组的异位子宫内膜组织、在位子宫内膜组织中的表达均低于对照组子宫内膜(P0. 05) ; P62蛋白在实验组的异位子宫内膜组织、在位子宫内膜组织中的表达高于对照组子宫内膜(P0. 05),但实验组的异位子宫内膜组织和在位子宫内膜组织中OPTN蛋白及P62蛋白的表达为异位组高于在位组,差异无统计学意义(P0.05)。[结论]1. EMs患者血清中OPTN蛋白高表达,P62蛋白低表达。2. EMs患者组织中OPTN蛋白、mRNA低表达,P62蛋白、mRNA高表达。3.各组织中自噬活性有所差异:正常子宫内膜组高于EMs患者在位内膜组高于异位子宫内膜组,自噬活性逐渐下降。4.本研究进一步证实了:自噬参与了 EMs的发生、发展,组织中OPTN低表达、P62高表达促进局部病灶发生、存活,血清中OPTN高表达、P62高表达可能是机体受到免疫、炎症反应等刺激后对自身的一种综合性保护机制。
[Abstract]:[objective] to detect the expression of Optineurin Optineurin Optineurin (OPTNN) P62 protein in serum and ectopic endometrium and eutopic endometrium (ovarian endometriosis cyst) of patients with endometriosis. To investigate the relationship between OPTNP 62 and endometriosis and to explore the role of autophagy related gene in the pathogenesis of endometriosis, and to find out the etiology of endometriosis by comparing the expression of OPTNP 62 in patients with endometriosis and normal control group, in order to understand the relationship between OPTNP 62 and endometriosis, and to explore the role of autophagy gene in the pathogenesis of endometriosis. To find new ideas for treatment. [methods] 1. Enzyme linked immunosorbent assay (Elisa) was used to detect the expression of OPTNN P62 protein. The expression of OPTNT P62 mRNA in ectopic endometrium, eutopic endometrium and normal endometrium of patients with EMs was detected by real-time fluorescence quantitative PCR assay (RT-PCR). The expression of OPTNP62 protein in ectopic endometrium, eutopic endometrium and normal endometrium of EMs patients was determined by Westren-Blot method. [results] the expression of 1.OPTN protein in serum of EMs patients was higher than that of normal control group (P 0. 0). The expression of P62 protein in serum of EMs patients was lower than that of normal control group. (05) .2. The expression of OPTN mRNA in ectopic endometrium and eutopic endometrium in experimental group was lower than that in control group. The expression of P62 mRNA in ectopic endometrium and eutopic endometrium in experimental group was higher than that in control group. However, the expression of OPTN mRNA and P62 mRNA in ectopic endometrium and eutopic endometrium in the experimental group was lower than that in the eutopic group, and there was no significant difference in the expression of OPTN mRNA and P62 mRNA in the ectopic endometrium of the experimental group. The expression of P0 in eutopic endometrium was lower than that in control group. The expression of P62 protein in ectopic endometrium and eutopic endometrium in experimental group was higher than that in control group. However, the expression of OPTN protein and P62 protein in ectopic endometrium and eutopic endometrium in experimental group was higher than that in eutopic group (P 0.05). [conclusion] 1. High expression of OPTN protein and low expression of P62 protein. 2. 2 in serum of EMs patients. The low expression of OPTN protein P62 protein and the high expression of P62 protein mRNA. 3. 3 in patients with EMs. The autophagy activity of normal endometrium group was higher than that of EMs group, and the autophagy activity of eutopic endometrium group was lower than that of ectopic endometrium group. This study further confirmed that autophagy was involved in the occurrence and development of EMs, the low expression of OPTN and the high expression of P62 promoted the occurrence and survival of local lesions. The high expression of OPTN in serum may be immune to the body. A comprehensive protective mechanism for itself after stimulation such as inflammation.
