自发性早产母体外周血转录组特征的研究

【摘要】:【目的】 (1)通过回顾性前瞻研究,分析自发性早产中母体外周血有核细胞转录组的分子特征; (2)通过对早产基因mRNA表达谱分析,筛选出差异表达基因,为研究早产分子机制提供线索。 【实验方法】 (1)基因表达谱分析:通过检测21例原始样本(10例自发性早产和11例足月妊娠)的mRNA表达谱,分析早产组和足月组间的基因表达特征,对其数据进行整合分析; (2)实时定量聚合酶链反应(RT-PCR):根据基因表达谱分析结果,挑选出13个上调差异基因,对其在另外46例独立样本(21例自发性早产和25例足月妊娠)中的表达水平进行验证。 【结果】 (1)基因表达谱分析结果:早产组中表达上调的基因241个,主要参与炎症、免疫应答、细胞增殖、DNA复制、DNA代谢、RNA剪切、RNA代谢、染色质组装等生物学过程;表达下调的基因690个,主要参与细胞稳态、信号转导、离子转运、核酸分解代谢等生物学过程。 (2)RT-PCR结果:选取差异最大的13个上调基因在独立样本中差异表达分析,,早产组中NF1表达水平高于足月组(P<0.05,P=0.008),差异具有统计学意义;早产组中CAMK2D表达水平高于足月组(P<0.05,P=0.028),差异具有统计学意义。 【结论】 (1)自发性早产的发生过程可能与相关基因激活和抑制有关; (2)NF1和CAMK2D基因表达异常可能参与早产的病理生理变化,有望成为预测早产的候选分子标志物。
[Abstract]:[purpose] 1) the molecular characteristics of nucleated cell transcriptome in maternal peripheral blood during spontaneous preterm delivery were analyzed by retrospective prospective study. 2) differential expression genes were screened by analyzing the mRNA expression profile of preterm birth genes, which provided clues for studying the molecular mechanism of preterm labor. [experimental method] (1) Analysis of gene expression profile: mRNA expression profiles of 21 cases of spontaneous preterm labor and 11 cases of term pregnancy were detected, the characteristics of gene expression between preterm and term groups were analyzed, and the data were integrated and analyzed. (2) Real-time quantitative polymerase chain reaction (RT PCR): according to the results of gene expression profiling, 13 up-regulated differentially expressed genes were selected and their expression levels were verified in another 46 independent samples (21 spontaneous preterm births and 25 term pregnancies). [results] Results of gene expression profile analysis: 241 up-regulated genes in preterm labor group were mainly involved in inflammation, immune response, cell proliferation, DNA replication, DNA metabolism, RNA shearing RNA metabolism, chromatin assembly, and 690 down-regulated genes. Mainly involved in cell homeostasis, signal transduction, ion transport, nucleic acid catabolism and other biological processes. RT-PCR results showed that the expression level of NF1 in preterm labor group was higher than that in term group (P < 0.05), and the difference was statistically significant. The expression of CAMK2D in premature delivery group was significantly higher than that in term group (P < 0.05). [conclusion] 1) the process of spontaneous preterm delivery may be related to the activation and inhibition of related genes; Abnormal expression of NF1 and CAMK2D may be involved in pathophysiological changes of preterm delivery and may be a candidate molecular marker for preterm delivery.
