黄体酮和LPS-RS对流产大鼠的作用及大鼠蜕膜中IL-1β、TNF-α的表达

【摘要】:目的:通过尾静脉注射脂多糖(LPS)建立大鼠流产的模型,用黄体酮及脂多糖拮抗剂LPS-RS干预,计算胚胎吸收率,并检测流产大鼠蜕膜组织中IL-1β、TNF-α因子蛋白表达情况,从而探讨感染性流产的发病机制及黄体酮和LPS-RS对感染引起流产的作用。 方法:将雌雄大鼠合笼,发现阴栓日为妊娠第0天,孕鼠随机分组,建立五组大鼠模型,实验一组为对照组,于孕7天尾静脉注射磷酸缓冲液(PBS)1ml/只,实验二组于孕7天尾静脉注射LPS1ml/只,实验三组于孕6-11天黄体酮肌注0.2ml/只,孕7天尾静脉注射LPS1ml/只,实验四组于孕7天尾静脉注射LPS-RS1ml/只后1小时再次尾静脉注射LPS1ml/只,实验五组于孕6-11天加黄体酮肌注0.2ml/只,孕7天尾静脉注射LPS-RS1ml/只后1小时再次尾静脉注射LPS1ml/只,分组即为对照组、LPS组(流产组)、LPS+黄体酮组,LPS+LPS-RS组,LPS+黄体酮+LPS-RS组,并于孕13天处死,取流产大鼠的蜕膜组织,计算胚胎吸收率,比较各组之间的差异,用western blot方法检测大鼠蜕膜组织IL-1β、TNF-α蛋白表达情况。 结果:五组胚胎吸收率分别为6.58%、24.79%、16.30%。9.89%、13.48%,实验二组(流产组)胚胎吸收率明显高于实验一组(对照组),成功建立大鼠流产模型,实验三组、实验四组、实验五组与实验一组比较,胚胎吸收率均高于实验一组,但无统计学意义,实验四组、实验五组与实验二组比较,胚胎吸收率均低于实验二组,有统计学意义,实验三组、实验四组、实验五组胚胎吸收率情况,两两比较无明显差异,无统计学意义。Western blot方法检测大鼠蜕膜组织的IL-1p蛋白表达情况的比较:实验一组、实验五组与实验二组(流产组)比较,实验二组大鼠蜕膜组织IL-1p表达上升,P0.05,有统计学意义,实验三组、实验四组、实验五组间两两比较大鼠蜕膜组织IL-1p表达未见明显差异,P0.05,无统计学意义。Western blot方法检测大鼠蜕膜组织的TNF-α表达情况:实验一组、实验五组与实验二组(流产组)比较,实验二组大鼠蜕膜组织TNF-α表达上升,P0.05,有统计学意义,实验三组、实验四组、实验五组间两两比较大鼠蜕膜组织TNF-α表达未见明显差异,P0.05,无统计学意义。 结论:脂多糖可引起妊娠大鼠流产,用LPS-RS及黄体酮处理后能一定程度上拮抗LPS导致的胚胎吸收。流产大鼠蜕膜组织中IL-1p、TNF-α蛋白表达增加,而黄体酮与LPS-RS合用后能使这两种细胞因子表达下调,提示激素合并抗炎治疗感染性流产效果更好。
[Abstract]:Objective: to establish a rat model of abortion by injecting lipopolysaccharide (LPS) into caudal vein. The embryo absorption rate was calculated with progesterone and lipopolysaccharide antagonist LPS-RS, and the expression of IL-1 尾 TNF- 伪 protein in decidua of aborted rats was detected. To explore the pathogenesis of infective abortion and the effect of progesterone and LPS-RS on infective abortion. Methods: the female and male rats were caged, and it was found that the day of Yin suppository was the first day of pregnancy. The pregnant rats were randomly divided into five groups. The experimental group was used as the control group. The rats were injected with phosphate buffer solution (PBS1ml / rat) at the end of the 7th day of gestation. LPS1ml/ was injected into the caudal vein on the 7th day of gestation, 0.2ml/ was injected intramuscularly by progesterone on the 6th to 11th day of gestation, LPS1ml/ was injected into the caudal vein on the 7th day of pregnancy, and LPS1ml/ was injected again one hour after LPS-RS1ml/ was injected into the caudal vein on the 7th day of gestation. The experimental five groups received intramuscular injection of 0.2ml/ with progesterone on 6-11 days of gestation, followed by re-injection of LPS1ml/ 1 hour after LPS-RS1ml/ was injected into the caudal vein on the 7th day of gestation. The rats were divided into two groups: control group (abortion group) and LPS-LPS-LPS-LPS-LPS-LPS-LPS-LPS-LPS-LPS-LPS-LPS-LPS-LPS-LPS-LPS-RS group. On the 13th day of gestation, the decidua tissues of aborted rats were taken, the embryonic absorption rate was calculated, and the differences among the groups were compared. The expression of IL-1 尾 -TNF- 伪 protein in decidua tissue of rats was detected by western blot method. Results: the embryo absorption rates of the five groups were 6.58 and 24.79 respectively. The embryo absorption rate of experimental group 2 (abortion group) was significantly higher than that of experimental group 1 (control group, successful establishment of rat abortion model, experimental group 3, experimental group 4, experimental group 5 and experimental group 1), and the embryo absorption rate of experimental group 2 (abortion group) was significantly higher than that of experimental group 1 (control group, experiment 3, experiment 4, experiment 5 and experiment 1). The embryo absorptivity of experiment group was higher than that of experimental group 1, but there was no statistical significance. Compared with experimental group 2, the embryo absorption rate of experimental group 4 and experimental group 5 was lower than that of experimental group 2, which had statistical significance. There was no significant difference in embryo absorption rate among the five experimental groups. There was no statistical significance. Western blot method was used to detect the expression of IL-1p protein in decidua of rats: group 1, group 5 and group 2 (abortion group) were compared with each other. The expression of IL-1p in decidual tissue of experimental group 2 was significantly higher than that of control group (P 0.05). There was no significant difference in the expression of IL-1p in decidua tissues between two groups. There was no statistical significance. Western blot method was used to detect the expression of TNF- 伪 in decidual tissues of rats: compared with experimental group 1, experimental group 5 and experimental group 2 (abortion group), the expression of TNF- 伪 in decidual tissue of rats was detected by Western blot. The expression of TNF- 伪 in decidua of experimental group 2 was significantly higher than that of control group (P 0.05). There was no significant difference in expression of TNF- 伪 between experimental group 3, experimental group 4 and experimental group 5 (P 0.05). Conclusion: lipopolysaccharide can induce abortion in pregnant rats. LPS-RS and progesterone can antagonize embryo absorption induced by LPS to some extent. The expression of IL-1pnTNF- 伪 protein in decidual tissue of abortive rats was increased, but progesterone combined with LPS-RS could down-regulate the expression of these two cytokines, suggesting that hormone combined with anti-inflammatory therapy is better in the treatment of infective abortion.
