围着床期小鼠子宫中巨噬细胞的变化及作用

【摘要】:目的观察小鼠子宫中胚胎着床部位与非着床部位巨噬细胞(Macrophage,Mφ)的分布差异,探讨其在胚胎着床过程中的作用。方法取真孕(n=30)和假孕小鼠(n=30)子宫组织,选择30只妊娠小鼠模型作为实验组,同时选择30只假孕小鼠模型作为对照组,在围着床期取两组小鼠子宫组织,采用免疫组化技术检测子宫组织中Mφ、诱导型一氧化氮合成酶(i NOS)、白血病抑制因子(LIF)的分布,分析比较子宫组织中胚胎着床部位和非着床部位Mφ、i NOS和LIF的分布差异及相关性。结果在小鼠子宫组织中,观察到实验组Mφ在D4.5(雌鼠交配后次日晨阴道见栓为D0.5)前主要分布子宫内膜,D4.5后主要分布于子宫外膜,对照组Mφ主要分布于子宫内膜;实验组i NOS在D4.5前主要分布子宫内膜,D4.5后主要见于子宫肌膜和外膜,对照组i NOS主要位于子宫内膜,且实验组Mφ和i NOS分布呈正相关。此外,实验组LIF显著表达于子宫内膜,对照组几乎不表达,二者间有统计学差异(P0.05);着床部位Mφ、i NOS和LIF明显高于非着床部位,其中Mφ、LIF比较有统计学差异(P0.05)。结论围着床期小鼠子宫组织中Mφ主要分布在子宫内膜及胚胎着床点,并伴有i NOS和LIF的表达,提示Mφ在胚胎着床中可能发挥重要作用。
[Abstract]:Objective to observe the distribution of Macrophagege M 蠁 (M 蠁) in mouse uterus and to explore the role of M 蠁 in embryo implantation. Methods the uterine tissues of the two groups were selected, 30 pregnant mice as experimental group and 30 pseudopregnant mice as control group. The uterine tissues of the two groups were taken during the period of surrounding bed. The distribution of M 蠁, inducible nitric oxide synthase (iNOS) and leukemia inhibitory factor (LIF) in uterine tissues were detected by immunohistochemical technique. The difference and correlation of M 蠁 NOS and LIF in embryo implantation and non-implantation were analyzed and compared. Results in the uterine tissues of mice, M 蠁 was mainly distributed in the endometrium before D4.5 (the vaginal thrombus was D0.5 in the morning after mating) in the experimental group, and mainly in the outer membrane after D4.5 in the control group, while in the control group, M 蠁 was mainly distributed in the endometrium. I NOS was mainly distributed in the myometrium and adventitia after D4.5 in the experimental group, and I NOS in the control group was mainly located in the endometrium, and the distribution of M 蠁 and I NOS in the experimental group was positively correlated. In addition, the expression of LIF was significantly higher in the experimental group than in the control group, and there was a significant difference between the two groups (P 0.05), and the M 蠁 I NOS and LIF in the implantation site were significantly higher than those in the non-implantation site, and there was a significant difference between the two groups (P 0.05). Conclusion M 蠁 mainly distributes in endometrium and embryo implantation site, accompanied by the expression of I NOS and LIF, suggesting that M 蠁 may play an important role in embryo implantation.
