卵巢衰老中细胞衰老现象初探

【摘要】:目的:第一部分:为了探究细胞衰老与卵巢衰老的关系,采用衰老相关的半乳糖苷酶(Senescence Associated b-galactosidase, SA-b-gal)染色方法观察在不同年龄阶段的成年C57BL/6J雌鼠卵巢组织细胞衰老现象的情况,探索在卵巢生理性衰老过程中,是否存在细胞衰老(cellular senescence)的现象,探索其在卵巢组织各类细胞中的分布,同时观察细胞衰老的程度随着年龄增长的变化; 第二部分:为了探究细胞衰老与卵泡早期发育的关系,本课题检测了新生及幼龄C57BL/6J雌鼠卵巢组织中细胞衰老的情况,探索在卵泡发育过程中细胞衰老现象的存在、分布以及变化。 通过以上两个方面阐明了细胞衰老与卵巢器官衰老、卵泡发育闭锁之间的关系。 方法:第一部分选取6周、12周、32周、40周、48周年龄阶段C57BL/6J雌性小鼠,检测各年龄段小鼠卵巢组织中衰老相关的半乳糖苷酶活性情况;第二部分选取2天、5天、7天及3周年龄阶段C57BL/6J雌性小鼠,检测各年龄段小鼠卵巢组织中衰老相关的半乳糖苷酶活性情况。 结果:通过衰老相关半乳糖苷酶活性检测法发现卵巢组织间质中,细胞衰老现象随着年龄的增长而增加,而在卵泡膜细胞中未见明显表达,颗粒细胞中未发现明显的增龄性趋势;在雌性小鼠出生后的卵泡早期发育过程中,未见明显细胞衰老现象出现在任何细胞中。 结论:在卵巢衰老过程中,间质细胞的衰老占据主要部分,并呈现随着年龄逐渐增长的趋势,在成年卵巢组织颗粒细胞细胞衰老现象未见明显增龄性趋势,而卵泡膜细胞未见衰老表现;在新生期卵泡早期发育过程中,未见细胞衰老现象。
[Abstract]:Objective: to investigate the relationship between cell senescence and ovarian senescence, senescence Associated b-galactosidase (SA-b-galgalase) staining was used to observe the aging of ovarian tissue cells in adult C57BL/6J female rats at different ages. To explore whether there is cellular senescence in the process of ovarian physiological senescence, to explore the distribution of cellular senescence in all kinds of ovarian cells, and to observe the change of cell senescence with age. The second part: in order to explore the relationship between cell senescence and early follicle development, we detected the cell senescence in ovarian tissue of newborn and young C57BL/6J mice, and explored the existence of cell senescence during follicular development. Distribution and variation. The relationship between cell senescence and ovarian organ senescence and follicular development atresia was clarified. Methods: in the first part, the aging related galactosidase activities in the ovarian tissues of C57BL/6J female mice were measured at the age of 6 weeks, 12 weeks, 32 weeks, 40 weeks and 48 weeks. In the second part, the senescence related galactosidase activity in ovarian tissues of C57BL/6J female mice of 2 days, 5 days, 7 days and 3 weeks was determined. Results: through the assay of senescence related galactosidase activity, it was found that cell senescence increased with age, but not in follicular membrane cells. There was no obvious aging trend in granulosa cells and no obvious cell senescence was found in any of the early follicles of female mice during the early development of postnatal follicles. Conclusion: in the process of ovarian senescence, the senescence of stromal cells occupies the main part and tends to increase with age, but there is no obvious aging trend in granulosa cells of adult ovarian tissue. However, there was no senescence in the follicular membrane cells and no senescence in the early development of the newborn follicles.
