IL-17A、IL-17F在卵巢上皮性肿瘤组织中的表达及其与微血管密度的关系

【摘要】:目的: 1.检测IL-17A和IL-17F在卵巢上皮性肿瘤及正常卵巢组织中的表达。 2.分析IL-17A和IL-17F与卵巢上皮性肿瘤各种临床病理参数和肿瘤微血管生成的关系,并初步探讨其可能机制。 3.评估IL-17A和IL-17F对患者预后的影响。 方法: 1.收集76例恶性卵巢上皮性肿瘤、10例交界性卵巢上皮性肿瘤、10例良性卵巢上皮性肿瘤和10例宫颈癌手术中切除的正常卵巢组织,采用免疫组织化学的方法,观察IL-17A和IL-17F的表达和分布特征。 2.采用免疫组织化学的方法对血管内皮细胞标志物CD105进行染色,计数恶性卵巢上皮性肿瘤中微血管的生成,探讨其与IL-17A、IL-17F表达的相关性。 3.统计分析IL-17A、IL-17F表达与临床资料的关系,并对随访资料进行生存分析。 结果: 1.恶性卵巢上皮性肿瘤组织中IL-17A、IL-17F呈高表达,且IL-17A与CD105(CD105标记血管内皮细胞计数MVD)的表达呈正相关(r=0.326, P0.05), IL-17F与CD105的表达呈负相关(r=-0.232,P0.05)。 2.IL-17A. IL-17F在卵巢癌组织中的表达与组织学分级、有无远处转移和有无淋巴结转移呈正相关(P0.05),而与患者年龄、肿瘤大小、病理分型、有无腹水无相关性(P0.05)。 3.卵巢癌组织中,IL-17A表达阳性组MVD值(4.95±1.91)明显高于阴性组(3.71±1.87)(P0.05), IL-17F表达阳性组MVD值(4.06-4-1.91)明显低于阴性组(5.22+2.23)(P0.05)。 4.单因素生存分析显示,组织学分级Ⅲ-Ⅳ级组平均生存时间较Ⅰ~Ⅱ级组平均生成时间短(P0.05);有远处转移组平均生存时间较无远处转移组平均生存时间短(P0.05);有淋巴结转移组平均生存时间较无淋巴结转移组平均生存时间短(P0.05);IL-17A高表达组平均生存时间较低表达组平均生存时间短(P0.01);IL-17F高表达组平均生存时间较低表达组平均生存时间短(P0.01)。多因素生存分析显示,IL-17A蛋白表达、IL-17F蛋白表达、组织学分级是影响恶性卵巢上皮肿瘤患者预后的独立因素(P0.05)。 结论: 1. IL-17A、IL-17F在卵巢上皮性肿瘤组织中的表达与肿瘤的恶性程度有关,肿瘤的恶性程度越高,IL-17A和IL-17F表达的阳性率越高。在卵巢上皮性肿瘤的发生、发展中起重要作用,可以作为临床判断卵巢上皮性肿瘤生物学行为有用的分子生物学指标。 2.IL-17A可发挥促卵巢癌微血管生成的作用,而IL-17F可发挥抑制微血管生成的作用,二者通过调节促血管生成和抗血管生成的平衡来促进肿瘤微血管的生成,且可作为预测卵巢上皮性肿瘤患者预后的独立危险因素。
[Abstract]:Objective: 1. The expression of IL-17A and IL-17F in epithelial ovarian tumors and normal ovarian tissues was detected. 2. To analyze the relationship between IL-17A and IL-17F and various clinicopathological parameters and microangiogenesis of ovarian epithelial tumors, and to explore its possible mechanism. 3. To evaluate the influence of IL-17A and IL-17F on the prognosis of patients. Methods: 1. Ten cases of borderline epithelial ovarian tumors and 10 cases of benign ovarian epithelial tumors and 10 cases of normal ovarian tissues were collected from 76 cases of malignant ovarian epithelial tumors and 10 cases of cervical cancer.
