孕期维生素A缺乏对大鼠子代肝脏脂质合成的影响

【摘要】:目的通过孕期维生素A(VA)缺乏大鼠模型,探讨孕期VA缺乏对子代肝脏组织中脂质合成的影响。方法建立大鼠孕期VA缺乏模型,分VA正常(VAN)、VA缺乏(VAD)、VA缺乏后补充(VAS)3组,检测子代血脂水平;肝脏中乙酰辅酶A羧化酶1(ACC1)、脂肪酸合成酶(FAS)、固醇调节元件结合蛋白(SREBP1)等脂质合成分子的mRNA表达变化;观察子代肝脏组织切片HE染色后脂滴沉积情况。结果 VAD组的HDL-C水平显著低于VAN组和VAS组(P0.05),而VAN组和VAS组之间差异无统计学意义(P0.05)。TG水平在3组间差异有统计学意义(P0.05)。VAD组ACC1、FAS、SREBP1mRNA表达水平显著增加。VAD组肝脏出现较多的脂滴沉积,细胞质中有部分脂滴空泡;而VAS组和VAN组肝细胞排列规则,未见明显脂滴沉积。结论孕期VA缺乏可诱发大鼠子代肝脏脂质合成通路的异常活化,造成质脂代谢紊乱。
[Abstract]:Objective to investigate the effect of vitamin A (VA) deficiency during pregnancy on lipid synthesis in liver tissue of offspring. Methods A model of VA deficiency during pregnancy was established in rats. The rats were divided into three groups: the normal VA group was supplemented with VASA deficiency group, and the blood lipid level of the offspring was detected. The changes of mRNA expression of lipids such as acetyl coA carboxylase 1 (ACC1), fatty acid synthase (FASA) and steroid regulatory element binding protein (SREBP1) in liver were observed. Results the level of HDL-C in VAD group was significantly lower than that in VAN group and VAS group, but there was no significant difference between VAN group and VAS group. There were some lipid droplets in the cytoplasm, but there was no obvious lipid droplet deposition in the liver cells of VAS group and VAN group. Conclusion during pregnancy, VA deficiency can induce abnormal activation of lipid synthesis pathway in rat offspring and lead to disorder of lipid metabolism.
