人阴道壁脱细胞基质的制备及其微纳结构分析

【摘要】:目的·探讨人阴道壁脱细胞基质的纳米/微米级结构特点,为生物材料制备仿生人工阴道提供参考数据。方法·对35例阴道壁脱垂的绝经后患者盆底重建手术中修剪下的阴道壁组织进行脱细胞处理,制备阴道壁脱细胞基质,并对脱细胞基质进行苏木精-伊红(H-E)染色、Masson染色、电镜扫描和Micro-CT扫描,采集并分析阴道壁脱细胞基质的大体形态及定量数据。结果·35例阴道壁标本脱细胞完全,基本无细胞残留。阴道壁脱细胞基质由互相交错的胶原纤维构成,为纳米级胶原纤维构成的双层纤维网状结构。胶原纤维呈束状排列,纤维束之间分布孔隙。阴道壁脱细胞基质上皮侧结构致密,为致密层;上皮下组织结构疏松,为疏松层。致密层和疏松层的纤维丝直径无显著差异[(82.4±9.3)nm vs(87.5±10.2)nm,P=0.432],纤维束宽度无显著差异[(67.6±9.3)μm vs(65.3±5.2)μm,P=0.634],疏松层的纤维束分离度较致密层大[(180.1±24.5)μm vs(118.2±23.0)μm,P=0.003],疏松层的孔隙率较致密层高[(77.1±4.2)%vs(66.6±2.8)%,P=0.002]。阴道前壁和阴道后壁的脱细胞基质的微纳结构参数没有显著差异。结论·人阴道壁脱细胞基质致密层和疏松层的纤维丝直径、纤维束宽度无显著差异,但致密层纤维束的孔隙率较疏松层小。
[Abstract]:Objective to study the nanometer / micron structure of human vaginal wall acellular matrix and to provide reference data for the preparation of biomimetic artificial vagina. Methods 35 postmenopausal patients with vaginal wall prolapse were treated with acellular tissue of vaginal wall trimmed during pelvic floor reconstruction, and the acellular matrix of vaginal wall was prepared, and the acellular matrix was stained by Masson staining with hematoxylin-eosin H-E staining. The morphological and quantitative data of acellular matrix of vaginal wall were collected and analyzed by electron microscopy and Micro-CT scanning. Results 35 cases of vaginal wall specimens were completely acellular and almost no cell residue. The acellular matrix of vaginal wall is composed of interlaced collagen fibers, which is a double fiber reticular structure composed of nanometer collagen fibers. The collagenous fibers were arranged in bundles, and the pores were distributed between the fibrous bundles. The acellular matrix epithelium of the vaginal wall is compact and the upper subcutaneous tissue is loose, which is the loose layer. There was no significant difference in fiber diameter between dense layer and loose layer [82.4 卤9.3)nm vs(87.5 卤10.2nmP0. 432], and there was no significant difference in the width of fiber bundle [67.6 卤9.3) 渭 m vs(65.3 卤5.2 渭 m P0. 634]. The separation degree of fiber bundle in loose layer was larger than that in dense layer [180.1 卤24. 5 渭 m vs(118.2 卤23. 0 渭 m P0. 003], and the porosity of loose layer was higher than that of dense layer [77.1 卤4.2)%vs(66.6 卤2. 8]. There was no significant difference in microstructural parameters of acellular matrix between anterior wall of vagina and posterior wall of vagina. Conclusion there is no significant difference in the diameter and width of fiber bundle between acellular matrix dense layer and loose layer of human vaginal wall, but the porosity of dense layer fiber bundle is smaller than that of loose layer.
