雄激素相关microRNA在PCOS患者卵巢颗粒细胞中的表达

【摘要】:目的:比较高雄激素多囊卵巢综合征患者(polycystic ovary syndrome,PCOS)组、非高雄激素PCOS患者组和正常对照组患者颗粒细胞中miR-21、miR-25、miR-132和miR-221的表达情况,探讨雄激素相关microRNA在高雄激素PCOS发病机制中的可能作用。 方法:选择2013年9月-2014年2月在中南大学湘雅医院生殖中心实施体外受精/卵胞浆内单精子显微注射-胚胎移植(IVF/ICSI-ET)治疗的女性不孕患者81例,其中高雄激素PCOS组(A组)23例,非高雄激素PCOS组(B组)27例,正常对照组(C组)31例。在取卵当日,回收废弃的卵泡液分离其中的颗粒细胞,进行原代培养及鉴定,采用荧光定量PCR法检测各组细胞中miR-21、miR-25、 miR-132和miR-221的表达差异情况,并与游离睾酮指数(FAI)进行相关性分析。 结果:1.人黄素化颗粒细胞体外培养24h后贴壁,48h后呈典型颗粒细胞样特征。2.A组miR-21和miR-25表达水平显著低于其他两组,差异有统计学意义(P0.05); miR-132和miR-221在三组患者中表达水平均无统计学差异(P0.05)。3.miR-21和miR-25的表达水平与FAI呈显著负相关(r=-0.331,P=0.003;r=-0.285,P=0.01),差异有统计学意义。 结论:高雄激素PCOS患者颗粒细胞中的miR-21和miR-25表达降低可能与高雄激素PCOS患者发病机制有关。
[Abstract]:Objective: to compare the expression of miR-21 miR-25 miR-132 and miR-221 in granulosa cells of patients with hyperandrogen polycystic ovary syndrome (PCOS), non-hyperandrogen PCOS patients and normal controls. To explore the possible role of androgen related microRNA in the pathogenesis of hyperandrogen PCOS. Methods: from September 2013 to February 2014, 81 female infertile women who were treated with IVF / ICSI-ETT at the Reproductive Center of Xiangya Hospital, Central South University, were selected, including 23 cases of hyperandrogen PCOS group (group A). No hyperandrogen PCOS group (n = 27) and control group (n = 31). The granulosa cells were isolated from the waste follicular fluid on the day of egg retrieval. The expression of miR-21 miR-25, miR-132 and miR-221 were detected by fluorescence quantitative PCR assay. Correlation analysis was made with free testosterone index (FAI). The result is 1: 1. Human luteinized granulosa cells were cultured in vitro for 24 hours. The expression of miR-21 and miR-25 in group A was significantly lower than that in the other two groups. There was no significant difference in the expression levels of miR-132 and miR-221 between the three groups. There was a significant negative correlation between the expression of miR-132 and miR-25 and FAI. Conclusion: the decreased expression of miR-21 and miR-25 in granulosa cells of patients with hyperandrogen PCOS may be related to the pathogenesis of hyperandrogen PCOS.
