体外受精对雌性小鼠骨骼肌11β-HSD1 H6pd及Nr3c1表达的影响

【摘要】:目的:研究IVF雌性小鼠骨骼肌基因11β-HSD1H6pd及Nr3cl的表达,探索IVF-ET子代成年后代谢性疾病发生的可能机制。 材料和方法: 1.利用C57BL/6J小鼠建立IVF及体内受精小鼠模型。 2.以IVF子代为研究对象,以体内受精子代为对照组,于3周(断乳期)、8周(性成熟期)、1.5年(老年期)取雌性小鼠骨骼肌。 3.通过qRT-PCR技术,检测3周、8周及1.5年龄时期雌性小鼠骨骼肌基因11β-HSD1H6pd及Nr3c1的表达。 4.通过western-blot技术,检测3周、8周及1.5年龄时期雌性小鼠骨骼肌上11β-HSD1蛋白的表达。 结果: 1.获IVF小鼠15只,体内受精小鼠15只。 2.IVF对出生后子代骨骼肌11β-HSD1H6pd及Nr3cl基因表达的影响 (1) qRT-PCR分析结果表明,IVF组小鼠子代与对照组之间骨骼肌组织上的11β-HSD1及H6pd基因表达从3周龄至老年期逐步出现差异,但两组间Nr3c1的表达各时期均无显著性差异。 (2)3周龄时11β-HSD1和H6pd的表达在两组之间差异无统计学意义;8周龄时,IVF组小鼠骨骼肌组织H6pd的表达显著性高于体内受精组,11β-HSD1的表达在两组之间无显著性差异;1.5年龄时,IVF组小鼠骨骼肌组织11β-HSD1和H6pd表达水平均较体内受精组显著性升高。Nr3c1的表达从3周龄至1.5年龄,两组间均无显著性差异。 3. IVF对出生后子代骨骼肌11β-HSD1蛋白表达的影响骨骼肌11β-HSD1蛋白的表达在两组间差异无统计学意义。 结论: 1. IVF技术可导致雌性小鼠子代骨骼肌的11β-HSD1和H6PD基因表达的改变。 2. IVF技术不影响雌性小鼠骨骼肌上的Nr3c1基因表达。
[Abstract]:Aim: to study the expression of 11 尾 -HSD1H6pd and Nr3cl in skeletal muscle of female IVF mice and to explore the possible mechanism of metabolic disease in adult IVF-ET offspring. Materials and methods: 1. The model of IVF and in vivo fertilization mice was established by using C57BL/6J mice. 2. The female skeletal muscles of female mice were collected from IVF offspring and spermatozoa in vivo as control group at 3 weeks (weaning stage 8 weeks (sexual maturity stage, 1.5 years old age). 3. The expression of skeletal muscle gene 11 尾 -HSD1H6pd and Nr3c1 in female mice at 3 weeks, 8 weeks and 1.5 years was detected by qRT-PCR technique. 4. The expression of 11 尾 -HSD1 protein on skeletal muscle of female mice at 3 weeks, 8 weeks and 1.5 years was detected by western-blot technique. Results: 1. Fifteen IVF mice and 15 fertilized mice in vivo were obtained. Effect of 2.IVF on expression of 11 尾 -HSD1H6PD and Nr3cl genes in skeletal muscle of postnatal generation The results of qRT-PCR analysis showed that the expression of 11 尾 -HSD1 and H6pd genes in skeletal muscle tissues of mice in the control group and in the control group was gradually different from 3 weeks to the old age, but there was no significant difference in the expression of Nr3c1 between the two groups at all stages. There was no significant difference between the two groups in the expression of 11 尾 -HSD1 and H6pd at the age of 3 weeks. The expression of H6pd in the skeletal muscle of the mice in the 8-week old group was significantly higher than that in the in vivo fertilization group. There was no significant difference between the two groups in the expression of 11 尾 -HSD1 and H6pd. The expression levels of 11 尾 -HSD1 and H6pd in skeletal muscle of mice in the control group were significantly higher than those in the in vivo fertilization group. The expression of Nr3c1 was from 3 weeks to 1.5 years old. There was no significant difference between the two groups. 3. Effect of IVF on the expression of 11 尾 -HSD1 protein in skeletal muscle after birth there was no significant difference between the two groups in the expression of 11 尾 -HSD1 protein in skeletal muscle. Conclusion: 1. The expression of 11 尾 -HSD1 and H6PD genes in skeletal muscle of female mice could be changed by IVF technique. 2. IVF technique did not affect the expression of Nr3c1 gene in skeletal muscle of female mice.
