卵巢生殖干细胞移植改善化疗小鼠卵巢功能的研究

【摘要】:目的探索小鼠卵巢生殖干细胞(OSCs)卵巢内移植是否可以改善化疗诱导的卵巢损伤。方法采用腹腔注射环磷酰胺和白消安建立化疗诱导的小鼠卵巢损伤模型,并随机分为模型组及移植组,同龄小鼠作为正常对照组。移植组在建模1周后,双侧卵巢分别移植OSCs。移植后观察小鼠动情周期;检测血清雌二醇(E2)和卵泡刺激素(FSH)浓度;测量卵巢重量及直径;卵巢组织切片观察卵巢结构和卵泡数目。结果 90%的模型组小鼠动情周期紊乱,70%移植组小鼠恢复正常的动情周期。移植组E2平均浓度(55.8±6.342)pg/ml高于模型组(34.1±4.643)pg/ml,差异有统计学意义(t=6.758,P0.05),而FSH浓度(28.3±1.433)ng/ml低于模型组(64.9±5.553)ng/ml,差异有统计学意义(t=-15.631,P0.05)。模型组卵巢呈萎缩状态,而移植组卵巢得到明显改善,形态、大小和正常对照组相似。移植组卵巢内各级卵泡数目均高于模型组,差异有统计学意义(t=11.851,P0.05),但仍低于正常对照组,差异亦有统计学意义(t=6.505,P0.05)。结论卵巢生殖干细胞卵巢内移植可以部分地修复化疗导致的卵巢损伤,从而改善卵巢功能。
[Abstract]:Objective to investigate whether intraovarian transplantation of mouse ovarian reproductive stem cell (OSCs) can improve ovarian injury induced by chemotherapy. Methods the ovarian injury model of mice induced by chemotherapy was established by intraperitoneal injection of cyclophosphamide and oxacol. The mice were randomly divided into model group and transplantation group, and the same age mice were used as normal control group. OSCs were transplanted into bilateral ovaries 1 week later in the transplantation group. After transplantation, the estrous cycle of mice was observed, serum estradiol E2) and follicle stimulating hormone (FSH) concentration were detected, ovarian weight and diameter were measured, ovarian structure and follicle number were observed in ovarian tissue sections. Results 90% of the mice in the model group had abnormal estrous cycle and 70% of the transplanted mice returned to normal estrous cycle. The average E _ 2 concentration in the transplantation group was 55.8 卤6.342)pg/ml higher than that in the model group (34.1 卤4.643) PG / ml, the difference was statistically significant (P < 0.05), while the concentration of FSH was 28.3 卤1.433)ng/ml lower than that in the model group (64.9 卤5.553ng / ml). The difference was statistically significant (t -15.631P 0.05). The model group showed atrophy, while the transplanted group got obvious improvement, shape, size and normal control group were similar. The number of ovarian follicles in the transplanted group was higher than that in the model group, and the difference was statistically significant (P 0.05), but still lower than that in the normal control group. The difference was also statistically significant. Conclusion Intraovarian transplantation of ovarian reproductive stem cells can partially repair ovarian damage induced by chemotherapy and improve ovarian function.
