缺氧通过刺激白介素8分泌促进宫颈癌细胞增殖

【摘要】:目的:探讨缺氧与白介素8对宫颈癌细胞的生存活力及凋亡是否具有调节作用及其可能的机制。 方法:免疫组化分析宫颈癌和宫颈炎组织中缺氧诱导因子(hypoxia inducible factor-1α,HIF-1α)、白介素8(interleukin8,IL-8)及其受体CXCR1和CXCR2的表达差异,采用磺酰罗丹明B(Sulforhodamine B,SRB)比色法及流式细胞术分析缺氧和白介素8对宫颈癌HeLa与SiHa细胞生存活力及凋亡的影响。结果:宫颈癌组织中HIF-1α、IL-8和CXCR1的表达水平明显高于宫颈炎 组织,缺氧可以刺激HeLa与SiHa细胞IL-8的分泌并增强其受体CXCR1和CXCR2的表达,重组人IL-8细胞因子可以促进HeLa与SiHa细胞的生存活力并降低凋亡率。在1%氧气条件下培养,HeLa与SiHa细胞的生存活力和凋亡水平增加,并且抗人IL-8中和性抗体可以部分阻断缺氧对HeLa和SiHa细胞活力的促进作用。 结论:缺氧可以诱导宫颈癌细胞分泌IL-8,,IL-8可以自分泌的方式增强宫颈癌细胞的生存活力,进而参与宫颈癌的发病。
[Abstract]:Objective: to investigate whether hypoxia and interleukin-8 can regulate the viability and apoptosis of cervical cancer cells and their possible mechanisms. Methods: the hypoxia-inducible factor-1 伪 -HIF-1 伪 in cervical carcinoma and cervicitis was analyzed by immunohistochemistry. The expression of IL-8 and its receptors CXCR1 and CXCR2 were different. The effects of hypoxia and interleukin-8 (IL-8) on the viability and apoptosis of cervical cancer HeLa and SIHA cells were analyzed by using sulfonyl rhodamine BSulforhodamine BHS-SRB colorimetry and flow cytometry. Results: the expression levels of HIF-1 伪, IL-8 and CXCR1 in cervical carcinoma were significantly higher than those in cervicitis. Hypoxia stimulated the secretion of IL-8 and enhanced the expression of CXCR1 and CXCR2 in HeLa and SIHA cells. Recombinant human IL-8 cytokines can promote the viability of HeLa and SIHA cells and decrease the apoptosis rate. The survival activity and apoptosis level of HeLa and SIHA cells cultured in 1% oxygen were increased. And anti-human IL-8 neutralizing antibody can partially block the effect of hypoxia on the activity of HeLa and SIHA cells. Conclusion: hypoxia can induce cervical cancer cells to secrete IL-8IL-8 to enhance the viability of cervical cancer cells in an autocrine manner. Then participate in the pathogenesis of cervical cancer.
