抑制URG4表达对宫颈癌小鼠的治疗作用及机制

【摘要】:目的:分析上调基因-4(URG4)在宫颈癌中的表达,并观察分析抑制URG4对宫颈癌的治疗作用及机制。方法:Western blot法分析URG4在宫颈癌组织及癌旁组织中的表达差异。选择16只雄性BALB/c裸鼠,喂养至6周龄后随机分为对照组(Control组)及观察组(Observation组),每组各8只。Control组小鼠于右侧背部皮下接种宫颈癌细胞(Si Ha cells)。Observation组小鼠背部皮下接种URG4-si RNA转染的宫颈癌细胞(URG4-si RNA Si Ha cells)。干预后4周处死小鼠,分析对比小鼠肿瘤生长情况及Wnt/β-catenin信号通路的改变。结果:URG4在人宫颈癌组织中呈现高表达,显著高于癌旁组织(P0.05)。干预结束后,观察组小鼠肿瘤组织内URG4表达量显著低于对照组(P0.05);体重明显高于对照组(P0.05);肿瘤组织重量显著低于对照组(P0.05)。观察组小鼠肿瘤组织内Ki67表达量显著低于对照组(P0.05)。观察组小鼠肿瘤组织内Cyclin D1、β-catenin及C-myc的表达均显著低于对照组,相比较差异均具有统计学意义(P0.05)。结论:URG4在人宫颈癌中呈现高表达,抑制URG4的表达可能通过抑制Wnt/β-catenin信号通路而发挥抗肿瘤效应。
[Abstract]:Aim: to investigate the expression of upregulation gene -4 urg _ 4 in cervical cancer, and to investigate the therapeutic effect and mechanism of inhibition of URG4 on cervical cancer. Methods the expression of URG4 in cervical carcinoma and paracancerous tissues was analyzed by Western blot. Sixteen male BALB / R / c nude mice were selected. After feeding to the age of 6 weeks, mice were randomly divided into control group (n = 8) and observation group (n = 8 in each group). The mice in each group were inoculated subcutaneously with cervix carcinoma cell line (Cervical carcinoma cell line) Si Ha cells transfected with URG4-si RNA on the back of mice in the right back of the mice in the control group and in the control group. The mice in the observation group were inoculated subcutaneously with URG4-si RNA transfected with URG4-si RNA. The mice were killed 4 weeks after intervention. The tumor growth and the changes of Wnt / 尾-catenin signaling pathway were analyzed and compared. Results the expression of 1% URG4 in human cervical carcinoma was higher than that in paracancerous tissue (P 0.05). After the intervention, the expression of URG4 in tumor tissues in the observation group was significantly lower than that in the control group (P 0.05), the body weight was significantly higher than that in the control group (P 0.05), and the weight of the tumor tissue was significantly lower than that in the control group (P 0.05). The expression of Ki67 in tumor tissues in the observation group was significantly lower than that in the control group (P 0.05). The expression of Cyclin D1, 尾 -catenin and C-myc in the tumor tissues of the observation group was significantly lower than that in the control group, and the difference was statistically significant (P 0.05). Conclusion the expression of 1: URG4 is highly expressed in human cervical carcinoma. Inhibition of the expression of URG4 may play an antitumor effect by inhibiting Wnt- 尾 -catenin signaling pathway.
