子宫颈鳞状细胞癌中Lgr5的表达及其临床意义

【摘要】：目的探讨富含酪氨酸重复序列G蛋白偶联受体(leucine-rich repeat-containing G protein-coupled receptor 5,Lgr5)在子宫颈鳞状细胞癌(cervical squamous cell carcinoma,CSCC)中的表达及意义。方法采用免疫组化EnVision两步法检测83例CSCC组织、56例癌旁正常组织及32例子宫颈上皮内病变(cervical intraepithelial neoplasia,CIN)组织中Lgr5蛋白表达,并应用RT-PCR检测13例CSCC及癌旁组织中Lgr5 mRNA的表达,分析Lgr5表达与CSCC临床病理特征的关系。结果 Lgr5在CSCC中的表达明显高于癌旁正常组织(P0.01)及CIN(P0.05)。Lgr5表达与肿瘤分化程度(P0.01)和浸润深度(P0.05)有关,与患者年龄、肿瘤大小、临床分期及淋巴结转移无关(P0.05)。Kaplan-Meier生存分析显示,Lgr5高表达者5年生存率显著低于Lgr5低表达者(P0.01)。多因素Cox比例风险回归模型分析显示,Lgr5的表达(P=0.027)可以作为判断CSCC患者预后的独立危险因素。结论 Lgr5的表达与肿瘤分化程度和浸润深度密切相关,可能参与CSCC的发生、发展,可作为CSCC患者预后判断的辅助指标。
[Abstract]:Objective to investigate the expression and significance of tyrosine rich repeat G protein-coupled receptor (leucine-rich repeat-containing G protein-coupled receptor 5 Lgr5) in cervical squamous cell carcinoma (cervical squamous cell carcinomonas carcinomas). Methods Immunohistochemical envision method was used to detect the expression of Lgr5 protein in 56 cases of adjacent normal tissues and 32 cases of cervical intraepithelial lesions (cervical intraepithelial neoplasia cin). RT-PCR was used to detect the expression of Lgr5 mRNA in 13 cases of CSCC and adjacent tissues. To analyze the relationship between the expression of Lgr5 and the clinicopathological features of CSCC. Results the expression of Lgr5 in CSCC was significantly higher than that in adjacent normal tissues (P0.01) and cin (P0.05) .Lgr5 expression was correlated with tumor differentiation (P0.01) and depth of invasion (P0.05). There was no correlation between clinical stage and lymph node metastasis (P0.05). Kaplan-Meier survival analysis showed that the 5-year survival rate of patients with high expression of Lgr5 was significantly lower than that of patients with low expression of Lgr5 (P0.01). Multivariate Cox proportional risk regression analysis showed that the expression of Lgr5 (P0. 027) could be used as an independent risk factor for the prognosis of CSCC patients. Conclusion the expression of Lgr5 is closely related to the degree of differentiation and depth of invasion of CSCC and may be involved in the occurrence and development of CSCC. It can be used as an auxiliary index to judge the prognosis of patients with CSCC.
