GATAD1在子痫前期中的表达及其意义

【摘要】：目的 :研究GATAD1在子痫前期患者胎盘组织中的表达及其意义。方法 :29例由美国Mayo Clinic妇产科临床医师收集的胎盘组织,其中早孕正常胎盘8例(分娩孕周为7~12周,平均为10.2周),晚孕正常胎盘14例(分娩孕周为35~41周,平均为39.5周),晚孕子痫前期胎盘7例(分娩孕周为31~41周,平均为34.6周)。采用RT-PCR技术、蛋白印迹(Western blot)法和免疫组化检测3组孕妇胎盘组织中GATAD1的m RNA与蛋白表达水平差异。结果:晚孕正常胎盘组织中GATAD1的m RNA与蛋白表达水平均显著高于早孕正常胎盘组织。子痫前期组患者胎盘组织中GATAD1的m RNA与蛋白表达水平均显著低于相应孕周正常妊娠组。结论:GATAD1表达的下降与子痫前期的发病相关。
[Abstract]:Objective: to study the expression and significance of GATAD1 in placenta of preeclampsia. Methods Twenty nine cases of placental tissue were collected by clinical doctors of gynecology and obstetrics at the Mayo Clinic in the United States. Among them, 8 cases were normal placenta of early pregnancy (7 weeks of gestation, averaging 10.2 weeks), 14 cases of normal placenta of late pregnancy (35 weeks and 41 weeks of gestation). The average gestational age was 39.5 weeks and the placenta of preeclampsia was found in 7 cases (the gestational age was 31 weeks, with an average of 34.6 weeks). The mRNA and protein expression levels of GATAD1 in placenta of pregnant women were detected by RT-PCR, Western blot and immunohistochemistry. Results: the mRNA and protein expression levels of GATAD1 in normal placenta of late pregnancy were significantly higher than those in normal placenta of early pregnancy. The expression of GATAD1 mRNA and protein in placenta of preeclampsia group was significantly lower than that of normal gestational group. Conclusion the decrease of the expression of Gata D1 is associated with preeclampsia.
