子宫内膜和卵巢两部位原发双癌中端粒长度检测及同源性研究

【摘要】:目的:分析宫腔子宫内膜样腺癌和卵巢子宫内膜样腺癌两部位癌组织中端粒长度是否相近,ER、PR及P53的表达是否相似,探讨两部位癌组织是否有同源性。 方法:收集子宫内膜和卵巢原发双癌患者的癌组织切除标本16例,子宫内膜癌组织切除标本43例,卵巢癌组织切除标本10例,采用荧光定量PCR法分别检测16例原发双癌患者两部位癌组织中端粒DNA的相对表达量,并采用免疫组化PV-6000二步法检测所有癌组织切除标本中免疫表型雌激素受体(Estrogen receptor ER)、孕激素受体(Progesterone receptor PR)及p53的表达。结果:(1)16例原发双癌患者,子宫内膜部位癌组织端粒相对长度(3.601±1.497)与卵巢部位癌组织端粒相对长度(3.71±1.556)相近,差异无统计学意义(t=1.423,P=0.175)。(2)原发双癌两部位癌组织ER蛋白免疫表型相同10例(62.5%),免疫表型不同6例(37.5%),差异无统计学意义(P=0.06);PR蛋白免疫表型相同9例(56.25%),免疫表型不同7例(43.75%),差异无统计学意义(P=0.09);P53蛋白免疫表型相同9例(56.25%),免疫表型不同7例(43.75%),差异无统计学意义(P=0.36)。(3)ER蛋白在单发性子宫内膜癌与原发双癌子宫内膜部位癌组织中(P=0.65)及单发性卵巢癌与原发双癌卵巢部位癌组织中(P=0.53)的表达差异均无统计学意义;PR蛋白在单发性子宫内膜癌与原发双癌子宫内膜部位癌组织中(P=0.48)及单发性卵巢癌与原发双癌卵巢部位癌组织中(P=0.42)的表达差异均无统计学意义;P53蛋白在单发性子宫内膜癌与原发双癌子宫内膜部位癌组织中(P=0.03)及单发性卵巢癌与原发双癌卵巢部位癌组织中(P=0.04)的表达差异均有统计学意义。 结论:(1)宫腔子宫内膜和卵巢两部位癌组织端粒长度相近,ER, PR及P53在两部位癌组织中的表达也相似。由此推断两部位癌组织可能有相同的组织来源,尚需进一步研究证实。 (2)ER、PR在单发性子宫内膜癌、卵巢癌中的表达与原发双癌相应部位癌组织中的表达相似,P53在单发性子宫内膜癌、卵巢癌中的表达与原发双癌相应部位癌组织中的表达均不同。因此,原发双癌与相应部位单发癌组织发生癌变的分子机制可能存在一定差异。
[Abstract]:Objective: to investigate whether the expression of telomere length in endometrial adenocarcinoma and ovarian endometrioid adenocarcinoma is similar to that of ERP PR and p53, and to explore whether there is homology between the two tissues. Methods: 16 specimens of carcinoma, 43 specimens of endometrial carcinoma and 10 specimens of ovarian cancer were collected from patients with primary carcinoma of endometrium and ovary. The relative expression of telomere DNA was detected by fluorescence quantitative PCR in 16 patients with primary double carcinomas. The expression of estrogen receptor (Estrogen receptor ER), progesterone receptor (Progesterone receptor PR) and p53 were detected by immunohistochemical PV-6000 two-step method. Results: (1) the relative length of telomere in endometrial carcinoma (3.601 卤1.497) and ovarian carcinoma (3.71 卤1.556) was similar to that in 16 patients with primary double cancer. The immunophenotype of ER protein was the same in 10 cases (62.5%) and different in 6 cases (37.5%). There was no significant difference in the immunophenotype of PR protein in 9 cases (56.25%), and in 7 cases (43.75%). There was no statistical significance (P0. 09). The immunophenotype of p53 protein was the same in 9 cases (56.25%), and the immunophenotype was different in 7 cases (43.75%). There was no significant difference (P0. 36). (3) in the expression of p53 protein in endometrial tissues of single endometrial carcinoma and primary double carcinomas (P0. 65), and between single ovarian carcinoma and primary double carcinoma. There was no significant difference in the expression of P0. 53 in the tissues of nests. There was no significant difference in the expression of PR protein between primary and single endometrial carcinomas (P0. 48) and primary ovarian cancer (P0. 42). There were significant differences in the expression of p53 protein between single endometrial carcinoma and primary double carcinoma of endometrium (P0. 03) and between single ovarian cancer and primary bicarcinoma of ovary (P0. 04). Conclusion: (1) the telomere length of endometrial and ovarian carcinoma is similar, and the expression of PR and p53 is similar. It is concluded that there may be the same tissue origin in the two sites of carcinoma, and further research is needed. (2) ER,PR in single endometrial carcinoma, The expression of p53 was similar to that in the corresponding tissues of primary double carcinomas, while the expression of p53 in ovarian cancer was different from that in the corresponding tissues of primary double carcinomas. Therefore, there may be some differences in the molecular mechanism of carcinogenesis between primary double carcinomas and corresponding single carcinomas.
