Notch信号通路配体Jagged1、Dll4在子痫前期胎盘组织中的表达

【摘要】:目的:探讨Notch信号通路配体Jagged1、Dll4在子痫前期胎盘组织中的表达及意义。方法:选取轻度子痫前期患者30例(轻度组)、重度子痫前期患者30例(重度组)、正常妊娠孕妇30例(对照组),采用免疫组化法检测其胎盘组织中Jagged1、Dll4的定位表达,采用荧光定量PCR技术和Western blot法分别检测胎盘组织中Jagged1、Dll4 mRNA及蛋白的表达。结果:Jagged1主要表达于血管内皮细胞和绒毛滋养层细胞,Dll4主要表达于血管内皮细胞。轻、重度组胎盘组织中Jagged1、Dll4 mRNA及蛋白的表达水平均低于对照组(P0.05),且重度组低于轻度组(P0.05);对照组胎盘组织中Jagged1与Dll4在mRNA和蛋白水平上的表达均呈正相关(r=0.806和0.808,P0.05),轻度组中两者表达的相关性减弱(r=0.675和0.560,P0.05),而在重度组中未发现两者的相关性。结论:Jagged1、Dll4在子痫前期胎盘组织中表达失衡,与子痫前期的发病关系密切。
[Abstract]:Aim: to investigate the expression and significance of Notch signal pathway ligand Jagged1,Dll4 in placenta of preeclampsia. Methods: 30 cases of mild preeclampsia (mild group), 30 cases of severe preeclampsia (severe group) and 30 cases of normal pregnant women (control group) were selected. The expression of Jagged1,Dll4 in placenta was detected by immunohistochemical method. The expression of Jagged1,Dll4 mRNA and protein in placenta was detected by fluorescence quantitative PCR and Western blot. Results Jagged1 was mainly expressed in vascular endothelial cells and chorionic trophoblast cells, and Dll4 was mainly expressed in vascular endothelial cells. Light, The expression of Jagged1,Dll4 mRNA and protein in placenta of severe group was lower than that of control group (P0.05), and that of severe group was lower than that of mild group (P0.05), and the expression of Jagged1 and Dll4 in placenta tissue of control group were positively correlated with mRNA and protein levels (r = 0.806 and 0.808%, P 0.05), respectively, and the expression of Jagged1 and Dll4 in placenta of severe group was significantly lower than that of mild group (P 0.05). The correlation between the two groups was decreased (r 0.675 and 0.560 P 0.05), but no correlation was found in the severe group. Conclusion the expression of Dll4 in preeclampsia placenta is out of balance, which is closely related to the pathogenesis of preeclampsia.
