孕妇尿酚类环境内分泌干扰物浓度检测的尿样收集方法比较

【摘要】:目的·探讨孕妇尿酚类环境内分泌干扰物检测的尿样收集方法,采用并比较2种方法对尿双酚A(BPA)、三氯生(TCS)与正壬基酚(4-n-NP)检测浓度的影响。方法·采用2种方法收集孕晚期尿样:导尿管尿袋收集(n=176),聚丙烯(PP)尿杯收集(n=642)。采用高效液相色谱 串联质谱(HPLC-MS/MS)法检测尿BPA、TCS及4-n-NP的浓度。验证实验采用PP材质尿杯、导尿管尿袋、低密度聚乙烯材质离心管及玻璃杯(阴性对照)留取同一妇女共5人的尿样。结果·导尿管尿袋收集的尿样BPA浓度几何均数为82.5 ng/mL(95%CI为71.4~95.4 ng/mL),比尿杯留取尿样的BPA浓度高63倍(几何均数1.3 ng/mL,95%CI为1.3~1.5 ng/mL);2种方法尿TCS、4-n-NP及肌酐浓度相似。验证实验的结果表明,相同个体的尿样,尿袋收集的尿BPA浓度远远高于其他3种方法。结论·尿酚类环境内分泌干扰物浓度检测的尿样收集,用PP材质尿杯优于导尿管尿袋收集,PP材质尿杯及低密度聚乙烯材质试管适合于流行病学人群研究。
[Abstract]:Objective to investigate the methods of urine sample collection for detection of environmental endocrine disruptors in urine of pregnant women, and to compare the effects of two methods on the detection of urinary bisphenol A (BPA), triclosan (TCS) and nonylphenol (4-n-NP). Methods two methods were used to collect urine samples in late pregnancy: urinary catheter bag collection (nm176), polypropylene (PP) urine cup collection (nm642). The concentrations of BPA,TCS and 4-n-NP in urine were determined by high performance liquid chromatography tandem mass spectrometry (HPLC-MS/MS). The urine samples of 5 women were collected from PP urine cup, urinary catheter bag, low density polyethylene centrifuge tube and glass cup (negative control). Results the geometric mean of BPA concentration in urine collected by urethral catheter was 82.5 ng/mL (95%CI was 71.4 卤95.4 ng/mL), which was 63 times higher than that in urine sample collected by urine cup (geometric mean 1.3 ng/mL,95%CI was 1.31.5 ng/mL). The results showed that the urine BPA concentration collected from the same sample was much higher than that of the other three methods. Conclusion PP urine cup is better than urethral catheter in collecting urine samples of environmental endocrine disruptor concentration of urine phenols. It is suitable for epidemiological population study to collect urethral cup made of PP and low density polyethylene (LDPE).
