甲状腺激素受体基因与卵巢癌顺铂耐药关系的研究

【摘要】:目的探讨甲状腺激素受体基因(THRB)是否为耐药相关基因或影响卵巢癌细胞耐药。方法合成特异性基因探针,利用实时荧光定量聚合酶链反应(qRT-PCR)技术显示细胞在激素作用下的基因表达差异。结果 THRB的表达在人卵巢癌耐顺铂SKOV3/DDP细胞中的表达量高于非耐药卵巢癌SKOV3细胞。结论甲状腺激素能促进人卵巢癌SKOV3细胞和人卵巢癌SKOV3/DDP耐顺铂细胞的增殖,THRB在卵巢癌耐药SKOV3/DDP细胞中的表达的量远高于一般卵巢癌SKOV3细胞,THRB对卵巢癌耐药起着非常重要的作用,可能是耐药相关基因,其机制有必要进一步探讨。
[Abstract]:Objective to investigate whether thyroid hormone receptor gene (THRB) is a drug resistance related gene or affects ovarian cancer cell resistance. Methods the specific gene probe was synthesized and the difference of gene expression in the cells under the action of hormone was demonstrated by real-time fluorescence quantitative polymerase chain reaction (qRT-PCR). Results the expression of THRB in cisplatin-resistant SKOV3/DDP cells was higher than that in non-drug-resistant ovarian cancer SKOV3 cells. Conclusion thyroid hormone can promote the proliferation of human ovarian cancer SKOV3 cells and human ovarian cancer SKOV3/DDP cisplatin resistant cells. The expression of THRB in drug-resistant SKOV3/DDP cells of ovarian cancer is much higher than that of normal ovarian cancer SKOV3 cells. THRB plays an important role in drug resistance of ovarian cancer. It may be a drug resistance related gene, and its mechanism needs to be further explored.
