纳米联合光动力治疗在卵巢癌中的应用

【摘要】:目的:探讨载药纳米粒联合光动力治疗应用于大鼠卵巢癌腹水瘤模型治疗的疗效。方法:制备包裹竹红菌乙素(HB)的聚氰基丙烯酸丁酯纳米粒(PBCA-NP)及建立Fischer 344大鼠卵巢癌腹水瘤模型。大鼠腹腔注射HB的DMSO溶液及HB-PBCA-NP溶液,观察不同药物剂型在大鼠体内分布,得到竹红菌乙素及竹红菌乙素纳米粒的药物动力学参数。将荷瘤大鼠随机分成4组:对照组未给予任何处理,手术组只接受手术治疗,实验组分别给予HB溶液及其纳米粒溶液,并接受手术及光动力治疗,观察生存时间。结果:纳米粒稳定,可用于体内实验。血清及肿瘤组织中,纳米粒组药物浓度达高峰的时间明显延迟。手术联合光动力治疗中位生存时间(95天和99天)比单纯手术(84天)长,可延长生存时间(P0.05)。结论:纳米粒可使药物在体内达到缓释,手术联合光动力治疗可延长生存时间。
[Abstract]:Objective: to investigate the therapeutic effect of drug loaded nanoparticles combined with photodynamic therapy on rat ovarian cancer ascites tumor model. Methods: Poly (butyl cyanoacrylate) nanoparticles (PBCA-NP) coated with (HB) were prepared and the ascites tumor model of ovarian cancer was established in Fischer 344 rats. Rats were injected intraperitoneally with DMSO solution and HB-PBCA-NP solution of HB. The pharmacokinetic parameters were obtained by observing the distribution of different drug formulations in rats. The rats were randomly divided into four groups: the control group did not receive any treatment, the operation group only received surgical treatment, the experimental group were given HB solution and nanoparticles solution, and received surgery and photodynamic therapy, and the survival time was observed. Results: the nanoparticles were stable and could be used for in vivo experiments. In serum and tumor tissue, the peak time of drug concentration in nanoparticles group was significantly delayed. The median survival time of operation combined with photodynamic therapy (95 days and 99 days) was longer than that of simple operation (84 days), and the survival time could be prolonged (P0.05). Conclusion: nanoparticles can make drug release in vivo, surgery combined with photodynamic therapy can prolong survival time.
