白藜芦醇下调Sirt1的表达对人卵巢癌Skov3细胞抑制作用的初步研究

【摘要】:目的: 通过观察白藜芦醇(Res)对人卵巢癌Skov3细胞的增殖抑制作用及其对Sirt1表达的影响,初步探讨Res对人卵巢癌Skov3细胞的生长抑制作用的相关机制,为卵巢癌的治疗提供实验和理论依据。 方法: 体外培养人卵巢癌Skov3细胞,用不同浓度(分别是0、5、10、20、40、80μg/ml)的Res作用于Skov3细胞24h、48h、72h,应用MTT方法检测不同浓度的药物对人卵巢癌Skov3细胞增殖的影响;应用RT-PCR和Western blot方法检测不同浓度的Res作用于人卵巢癌Skov3细胞24h后其Sirt1mRNA和蛋白表达水平的变化。 结果: 1.不同浓度的Res(0、5、10、20、40、80μg/ml)作用于人卵巢癌Skov3细胞不同时间(24h、48h、72h)后,,人卵巢癌Skov3细胞的生长受到抑制,且抑制作用随时间和浓度的增加而增强,组与组之间的比较及组与对照组间的比较,差异均有统计学意义(P0.05); 2.不同浓度的Res(0、5、10、20、40、80μg/ml)作用于人卵巢癌Skov3细胞24h,随着药物浓度的增加,人卵巢癌Skov3细胞中Sirt1mRNA的表达逐渐下调(P0.05); 3.不同浓度的Res(0、5、10、20、40、80μg/ml)作用于人卵巢癌Skov3细胞24h,随着药物浓度的增加,人卵巢癌Skov3的Sirt1蛋白的表达逐渐下调(P0.05)。 结论: 1. Res能抑制人卵巢癌Skov3细胞的增殖,且抑制作用存在时间浓度依赖关系。 2. Res可能是通过下调Sirt1的表达发挥对人卵巢癌Skov3细胞的抑制作用。
[Abstract]:Aim: to investigate the inhibitory effect of resveratrol (Res) on the proliferation of human ovarian cancer Skov3 cells and the expression of Sirt1, and to explore the mechanism of Res on the growth inhibition of human ovarian cancer Skov3 cells. To provide experimental and theoretical basis for the treatment of ovarian cancer. Methods: human ovarian cancer Skov3 cells were cultured in vitro and treated with different concentrations of Res (0 ~ (51010) ~ 20 ~ 4080 渭 g/ml) for 24 h ~ 48 h ~ 72 h. The effects of different concentrations of drugs on the proliferation of human ovarian cancer Skov3 cells were detected by MTT method. RT-PCR and Western blot methods were used to detect the expression of Sirt1mRNA and protein in human ovarian cancer Skov3 cells treated with different concentrations of Res for 24 hours. Results: 1. The growth of human ovarian cancer Skov3 cells was inhibited by different concentrations of Res (0H5, 1020, 4080 渭 g/ml) at different time (24 h, 48 h, 72 h), and the inhibitory effect increased with the increase of time and concentration. The differences between the two groups and between the two groups were statistically significant (P0.05). 2. The expression of Sirt1mRNA in human ovarian cancer Skov3 cells decreased gradually with the increase of drug concentration (P 0.05). The expression of Sirt1 protein in human ovarian cancer Skov3 cells decreased gradually with the increase of drug concentration (P05). Conclusion: 1. Res can inhibit the proliferation of human ovarian cancer Skov3 cells in a time and concentration dependent manner. 2. Res may play an inhibitory role on human ovarian cancer Skov3 cells by down-regulating the expression of Sirt1.
