siRNA沉默MACC1基因表达对宫颈癌细胞增殖及侵袭的影响

【摘要】:目的探讨siRNA沉默结肠癌转移相关基因1(MACC1)表达对Hela宫颈癌细胞增殖及侵袭的影响。方法通过脂质体将siRNA MACC1及siRNA NC转入Hela细胞中,RT-PCR及western blot检测转染效果,MTT法检测细胞活力,流式细胞术检测细胞周期,Transwell法检测细胞侵袭能力,western blot检测细胞周期蛋白D1(cyclin D1)、cyclin E,基质金属蛋白酶2(MMP2)、MMP9蛋白表达及细胞外调节蛋白激酶(ERK)磷酸化水平。结果与siRNA NC组比较,siRNA MACC1组中MACC1蛋白及mRNA表达量降低(P0.01),细胞活力及侵袭能力下降(P0.01),细胞周期阻滞在G1期,cyclin D1、cyclin E、MMP2、MMP9及p-ERK表达量下调(P0.01)。结论siRNA MACC1能显著的抑制宫颈癌Hela细胞增殖与侵袭,可能与下调ERK磷酸化水平有关。
[Abstract]:Objective to investigate the effect of siRNA silencing the expression of metastasis associated gene 1 (MACC1) on the proliferation and invasion of Hela cervical cancer cells. Methods siRNA MACC1 and siRNA NC were transferred into Hela cells by liposome. Transfection effect was detected by RT-PCR and western blot. Cell viability was detected by MTT assay, cell cycle was detected by flow cytometry, and cell invasion ability was detected by Transwell assay. The expression of cyclin D1), cyclin E, matrix metalloproteinase 2 (MMP2), the expression of MMP9 protein and the phosphorylation of extracellular regulated protein kinase (ERK) were detected by western blot. Results compared with siRNA NC group, the expression of MACC1 protein and mRNA in siRNA MACC1 group was decreased (P0.01), cell viability and invasiveness were decreased (P0.01), cell cycle was blocked in G1 phase, and cyclin D1 cyclin EmMP2 was found in siRNA MACC1 group. The expression of MMP9 and p-ERK was down-regulated (P0.01). Conclusion siRNA MACC1 can significantly inhibit the proliferation and invasion of cervical cancer Hela cells, which may be related to the down-regulation of ERK phosphorylation.
