宫颈癌组织中maspin、AKT、p-AKT表达变化及其与肿瘤微淋巴管密度的相关性

【摘要】:目的观察宫颈癌组织中maspin、蛋白激酶B(AKT)、磷酸化蛋白激酶B(p-AKT)的表达变化,并分析其与肿瘤微淋巴管密度(LMVD)的相关性。方法收集127例份宫颈石蜡组织标本,其中慢性宫颈炎组织20例份(炎症组)、CINⅠ级组织20例份(CIN1组)、CINⅡ级组织20例份(CIN2组)、CINⅢ级组织20例份(CIN3组)、宫颈癌组织47例份(宫颈癌组,其中无淋巴结转移32例为宫颈癌1组、有淋巴结转移15例为宫颈癌2组)。采用免疫组化SP法检测各组maspin、AKT、P-AKT,测算肿瘤微淋巴管密度(LMVD)。结果宫颈癌1组、宫颈癌2组maspin蛋白阳性表达比例低于其余各组(P均0.05)。宫颈癌1组、宫颈癌2组AKT、p-AKT阳性表达比例高于其余三组(P均0.05)。宫颈癌2组p-AKT阳性表达比例高于宫颈癌1组(P0.05)。宫颈癌组织中maspin与AKT表达呈负相关关系(r_s=-0.472,P=0.001),maspin与p-AKT表达也呈负相关关系(r_s=-0.598,P=0.000)。宫颈癌1组、宫颈癌2组LMVD分别为(19.44±2.95)、(23.24±2.46)支/mm~2,淋巴结转移与LMVD呈正相关关系(r_s=0.44,P=0.000)。宫颈癌组织中LMVD与maspin表达呈负相关关系(r_s=-0.554,P=0.000),LMVD与p-AKT表达呈正相关关系(r_s=0.39,P=0.007)。结论宫颈癌组织中maspin表达下调,AKT、p-AKT表达上调,且maspin表达与AKT、p-AKT表达均呈负相关关系;宫颈癌组织中LMVD与maspin表达呈负相关关系,LMVD与p-AKT表达呈正相关关系。maspin表达下调可能参与了宫颈癌的发生发展,机制可能与AKT通路异常激活、促进肿瘤微淋巴管生成有关。
