肠凝集素1稳定表达对裸鼠人子宫内膜癌Ishikawa细胞移植瘤生长的影响

【摘要】:目的 :探讨肠凝集素1(intelectin-1,ITLN-1)稳定表达对人子宫内膜腺癌Ishikawa细胞裸鼠移植瘤生长的影响。方法 :16只裸鼠随机分为2组,分别皮下接种感染慢病毒GV358(携带有ITLN-1基因的慢病毒)的Ishikawa细胞(ITLN-1组)和感染空载体慢病毒(仅含有空载体的慢病毒)的Ishikawa细胞(对照组),建立裸鼠人子宫内膜癌Ishikawa细胞移植瘤模型。测量小鼠皮下肿瘤体积,绘制肿瘤生长曲线,剥离肿瘤后称取肿瘤的质量,计算肿瘤抑制率;肿瘤组织切片行HE染色观察病理学形态;免疫组织化学法检测肿瘤组织中p53、环氧合酶2(cyclooxygenase-2,Cox-2)和Bcl-2蛋白的表达情况。结果 :与对照组相比,ITLN-1组小鼠移植瘤的生长受到明显抑制(P0.01);免疫组织化学结果显示,ITLN-1组中野生型p53蛋白的表达水平明显上调(P0.01),而Cox-2和Bcl-2蛋白的表达水平明显下调(P值均0.01)。结论 :ITLN-1过表达能抑制裸鼠人子宫内膜癌移植瘤的生长,发挥抗肿瘤作用。
[Abstract]:Aim: to investigate the effect of stable expression of lectin 1 (intelectin-1,ITLN-1) on the growth of human endometrial adenocarcinoma Ishikawa cells in nude mice. Methods: sixteen nude mice were randomly divided into two groups. Ishikawa cells infected with lentivirus GV358 (lentivirus carrying ITLN-1 gene) (ITLN-1 group) and Ishikawa cells infected with empty vector lentivirus (lentivirus only) were inoculated subcutaneously (control group). Human endometrial carcinoma (Ishikawa) transplanted tumor model was established in nude mice. The tumor volume was measured, the tumor growth curve was drawn, the tumor mass was removed after the tumor was stripped, the tumor inhibition rate was calculated, and the pathological morphology was observed by HE staining. The expression of p53, cyclooxygenase 2 (cyclooxygenase-2,Cox-2) and Bcl-2 protein were detected by immunohistochemistry. Results: compared with the control group, the growth of transplanted tumor in ITLN-1 group was significantly inhibited (P0.01). Immunohistochemical results showed that the expression of wild-type p53 protein was up-regulated in ITLN-1 group (P0.01), while the expression of Cox-2 and Bcl-2 protein was down-regulated (P0.01). Conclusion: overexpression of ITLN-1 can inhibit the growth of human endometrial carcinoma xenografts in nude mice and play an anti-tumor role.
