PPARγ在绒毛组织中的表达与早期自然流产的关系
[Abstract]:Spontaneous abortion is the most common disease of pregnancy. According to statistics, 31% of the embryos have spontaneous abortion after implantation, 80% of which are early abortion [1], its etiology is complex and diverse, and the exact pathogenesis is still unclear. In recent years, peroxisome proliferator-activated receptor- 纬 (PPAR 纬), a member of the nuclear receptor transcription factor superfamily, has been found to play an indispensable role in placenta formation and development during pregnancy. Abnormal expression in various pregnancy-related diseases is expected to be a potential therapeutic target for the treatment and prevention of adverse pregnancy outcomes in future research. Objective: to study the differential expression of PPAR 纬 in villi of normal early pregnancy and early spontaneous abortion, and to analyze the possible relationship between PPAR 纬 and early spontaneous abortion. Methods: the expression of PPAR 纬 protein in the villi of 40 normal early pregnant women (control group) and 40 early spontaneous abortion patients (experimental group) was detected by two step immunohistochemical method (Power Vision,PV). Western Blot,RT-PCR technique was used to detect the expression of PPAR 纬 protein and gene in chorionic villi of the two groups, and to analyze whether there were significant differences between the two groups. There was no complication of pregnancy and complications in the two groups. The history of adverse pregnancy and childbirth was denied. The patients did not take steroid drugs 1 month before operation. There was no significant difference in clinical characteristics between the two groups. Results: 1. Immunohistochemical staining showed that PPAR 纬 protein was mainly expressed in trophoblast cells and extravillus trophoblastic cells in the two groups, and the expression level of PPAR 纬 in the experimental group was higher than that in the control group (P0.05). 2.Western Blot analysis showed that the expression of PPAR 纬 protein in the chorionic villi of the experimental group was higher than that in the control group (P0.01). 3.RT-PCR analysis showed that the expression of PPAR 纬 gene in the chorionic villi of the experimental group was higher than that in the control group. The difference was significant (P0.01). Conclusion: 1.PPAR 纬 protein is mainly expressed in cytotrophoblast cells and extravillous trophoblast cells in villi of early pregnancy and spontaneous abortion. The expression of 2.PPAR 纬 protein and mRNA in villi of early spontaneous abortion patients was significantly higher than that of normal early pregnancy, which may be closely related to the occurrence of spontaneous abortion.
