辣椒碱对人宫颈癌细胞Hela和C-33A的抗肿瘤活性

【摘要】：目的:研究辣椒碱对Hela(HPV+)和C-33A(HPV-)细胞生长增殖和凋亡的影响,探讨辣椒碱对宫颈癌细胞的抑制作用与相关机制。方法:选取HPV(+)的Hela和HPV(-)的C-33A宫颈癌细胞株为研究对象。CCK-8法检测细胞活性,流式细胞术检测辣椒碱对细胞生长的抑制作用、细胞周期和对细胞凋亡的影响;q PCR和Western blot法检测细胞凋亡相关基因m RNA和蛋白表达。结果:辣椒碱对宫颈癌Hela和C-33A细胞增殖抑制呈浓度和时间依赖性;辣椒碱能显著促进两种细胞凋亡,对Hela细胞的促凋亡效果更明显。辣椒碱处理后,两种细胞的细胞色素C(cytchrome C)和caspase3表达量显著增加,Bax表达量增加,Bcl-2表达量降低。结论:辣椒碱可显著抑制宫颈癌Hela和C-33A细胞增殖和促进细胞的凋亡,其机理可能是通过诱导细胞内活性氧类物质(ROS)增加、细胞色素C和Caspase3表达量增加等途径,最终诱导细胞凋亡。
