TET1在子宫内膜病变中的表达及其对癌细胞增殖和迁移能力的影响

【摘要】:目的探讨不同类型子宫内膜病变中TET1的表达水平和干扰TET1蛋白表达对子宫内膜癌细胞系Hec-1a和Ishikawa细胞生物学行为的影响。方法免疫组织化学检测组织中TET1的表达;应用干扰TET1表达的siRNA转染Hec-1a和Ishikawa细胞,以仅加入转染试剂的细胞作为阴性对照;Western blotting检测转染效率;SRB法检测细胞增殖能力改变;Transwell侵袭试验检测细胞侵袭能力改变;划痕试验检测细胞迁移能力改变;流式细胞术检测细胞周期改变。结果子宫内膜组织中TET1的表达随病理类型进展而升高;干扰组TET1蛋白表达水平明显低于对照组;干扰TET1表达后子宫内膜癌细胞增殖、迁移和侵袭能力均受到不同程度抑制,同时出现G1/S期细胞周期阻滞。结论子宫内膜癌及子宫内膜增生性病变组织中TET1表达水平明显高于正常对照组织;干扰TET1表达后子宫内膜癌细胞的增殖、迁移和侵袭能力下降,且出现G1/S期细胞周期阻滞,提示TET1在子宫内膜癌的增殖和转移过程中发挥重要作用。
[Abstract]:Objective to investigate the effects of TET1 expression and interfering TET1 protein expression on the biological behavior of Hec-1a and Ishikawa cells in different types of endometrial lesions. Methods Immunohistochemical method was used to detect the expression of TET1 in tissues, Hec-1a and Ishikawa cells were transfected with siRNA which interfered with TET1 expression,; Western blotting was used as negative control to detect the efficiency of; Western blotting, and SRB assay was used to detect the change of cell proliferation ability. Transwell invasion test was used to detect the change of cell invasion ability, scratch test was used to detect the change of cell migration ability, and flow cytometry was used to detect the change of cell cycle. Results the expression of TET1 in endometrial tissue increased with the progress of pathological type, and the expression level of TET1 protein in interference group was significantly lower than that in control group. After interfering with the expression of TET1, the proliferation, migration and invasion of endometrial cancer cells were inhibited in varying degrees, and the cell cycle arrest of G _ 1 鈮,
